Research ContextGinkgo biloba extract is a medicinal active ingredient extracted from the dried leaves of ginkgo trees.Its active ingredients are flavonoids and terpene lactone compound,and a small amount of amino acids,sugars,vitamins and minerals.Ginkgo biloba extract is mainly used in the clinical treatment of cardiovascular disease,are used in cosmetics.It has free radical scavenging,antioxidant,anti-PAF(platelet activating factor),anti-viral effects.Rhagadia manus is a common clinical disease in Department of dermatology.Its incidence rate is higher.It's clinical manifestation was the back of the hand,fingers and heel skin rough and cracking.t is associated with severe cases of bleeding and pain,while susceptible to bacterial infections.This disease is more common in the elderly and women.Due to frequent exposure to chemical or mechanical substance,especially in the cold winter weather,the skin and sweat glands to reduce dry skin,resulting in skin thickening and skin loses its elasticity.So when the hand and foot when prone to chapped.Gel ointment(cataplasm)is water soluble polymer materials as the main substrate,while adding drugs,by mixing together,coating,cutting etc.process,thus prepared.Compared with the dosage form,cataplasm has the advantages of high bioavailability,avoiding first pass effect,less dosage,high drug loading amount,strong target resistance,no irritation,and convenient use.The purpose of this study is based on Ginkgo biloba extract as a model drug and Gel ointment as as a carrier,which is made of suitable percutaneous administration preparations for clinical purpose is to provide safe and effective treatment of rhagadia manus and frostbite.Main contents1.The use of single factor and prescription transdermal in vitro experiments,Ginkgo biloba extract gel ointment formulation was optimized.2.The quality of the gel paste of Ginkgo biloba extract was evaluated while establishing the method which content of ginkgo flavonol glycosides(including quercetin,kaempferol and isorhamnetin)can detected.3.Different forms of Ginkgo biloba extract were compared by which was measured in vitro percutaneous transdermal flux.4.Ginkgo biloba extract for the efficacy of rat model of skin chapped and preliminary efficacy of mice ears frostbite model and irritation and acute toxicity were observed,for providing theoretical and experimental basis for treating rhagadia manus and frostbite in clinic.Methods1.To optimize prescription of gel paste of Ginkgo biloba extract The prescription of gel paste of Ginkgo biloba extract was optimized,including adjustment of the viscosity and selection of transdermal penetration enhancers.In the aspect of adjustment of the viscosity of cataplasms by changing the content of tartaric acid,it was compared with holding viscosity of cataplasms with different content of tartaric acid.In the selection of transdermal permeation enhancers,a modified Franz diffusion pool were used to study the effects of 3% azone,3% azone + 3% borneol and 3% azone + 3% propylene glycol in vitro percutaneous absorptionon of ginkgo flavonol glycosides of gel paste.2.Determination of flavonoids of Ginkgo biloba extract in the gel pasteThe method of HPLC is adopted to establish a method for determination of content of ginkgo flavonol glycosides(including quercetin,kaempferol and isorhamnetin).Referring to the pharmacopoeia requirements,containing paste volume,shaping and adhesion should be checked in gel paste of Ginkgo biloba extract.3.In vitro percutaneous penetration effect of Ginkgo biloba extract in gel pasteA modified Franz diffusion pool and mouse skin in vitro as permeable membranes were used to study the effects of the gel paste,cream and alcohol solutions through the skin by the establishment of a t method of percutaneous penetration experiment in gel paste of Ginkgo biloba extract.4.Evaluate the pharmacodynamics and safety of developed gel paste of Ginkgo biloba extract.The chapped skin model of rat and the ear frostbite model of mouse were used to investigate the therapeutic effect of gel paste of Ginkgo biloba extract on the rat model of skin chapped,as well as preliminary curative effect on mouse ear frostbite model.At the same time,the animal skin irritation and acute toxicity were also studied.Results1.In the aspect of adjustment of the viscosity of cataplasms,when the 100 g matrix containing 0.2004 g acid,the viscosity of gel paste of Ginkgo biloba extract is the best.In the selection of transdermal permeation enhancers,the cataplasms containing azone and propylene glycol is superior to others containing azone and others containing azone and borneol.3% azone and 3% propylene glycol can be used as penetration enhancers in gel paste of Ginkgo biloba extract.2.By researching methodology,the determination method of ginkgo flavonol glycosides is exclusive,high precision and high recovery rate.And it can be used for the determination of content of ginkgo flavone glycol glycosides accurately.3.By researching methodology,the analysis method of percutaneous permeation experiment of gel paste of Ginkgo biloba extract has good specificity,precision and recovery.It can be used to determine the content of total flavonoids of Ginkgo biloba in the osmotic solution.Comparison of gel paste,cream and alcohol solutions through the skin,the results show that,effect of gel paste of Ginkgo biloba extract through the skin is better than the other two forms.4.Skin irritation,acute toxicity and preliminary pharmacological studies show that the homemade gel paste of Ginkgo biloba extract is non-toxic,nonirritant and has good safety.It has a better therapeutic effect on a rat model of chapped skin.And It can relieve the mice ears frostbite model caused by swelling and inflammation.The method is simple and rapid,and it can be used to determine the content of Ginkgo biloba extract accurately.The gel paste is feasible in preparation technique,stable and controllable in quality and can improve transdermal penetration and increased the Ginkgo biloba extract amount retain in the skin significantly.Pharmacodynamic studies have shown that gel paste of ginkgo biloba extract has good resistance to the effect of cracked and is preliminary treatment for frostbite.The results of skin irritation and acute toxicity tests show that gel paste of Ginkgo biloba extract was non-toxic,no stimulation and good safety.
