Objective:1.To observe the effect of echinacoside from Cistanche on survival time of mice in hypoxia tolerance experiment.2.To observe the effect and possible mechanism of echinacoside on rats with high altitude pulmonary hypertension.Method:1.Compared the surivival time of mice in hypoxia tolerance experiment.2.Keeping rats in simulated 5000m altitude environment for four weeks to establish rat model of pulmonary hypertension,administered Rhodiola(1.78ml/kg)and echinacoside(12.5mg/kg,25mg/kg,50mg/kg)for 4 weeks.Test the mPAP and RVSP directly,RVHI was calculated.Observe the morphological changes of the heart tissue and pulmonary artery under light microscope.The levels of IL-6,TNF-?,NO content,NOS activity in lung were measured by ELSA and ET-1 content were measured by Radioimmunoassay.Detected the expression of VEGF,TGF-?1 protein in lung tissue were measured by Immunohistochemical and Western Blot.Detected the expression of VEGF,TGF-?1 mRNA in lung tissue were measured by RT-PCR.Result:1.The survival time was extened both in echinacoside dose groups and Rhodiola group(p<0.01).2.Echinacoside dose groups and Rhodiola crenulata administration can reduce the mean pulmonary arterial pressure,right ventricular systolic pressure and right ventricular hypertrophy;inhibit rat pulmonary vascular thickening,inflammatory cell infiltration and myocardial cell hypertrophy;reduce IL-6,TNF-?,ET-1(p<0.01)content while increase NO content and activity of NOS(p<0.01)in lung.Inhibit the VEGF,TGF-?1 protein and mRNA expression in lung(p<0.01).Conclusion:1.Echinacoside had benefit effect on the high-altitudue hypoxia.2.Keeping rats in simulated 5000m altitude environment can successfully establish rat model of pulmonary hypertension.3.The effect of echinacoside on rats with high altitude pulmonary hypertension may be related to the antihypoxia capacity,reduction of pulmonary bemodynamics,improve the pulmonary vascular remodeling and cardiac hypertrophy,anti-inflammatory,dilation of blood vessels,inhibit vascular contraction and the expression of VEGF,TGF-?1 in lung tissue.
