Objective: To investigate the effect and mechanism of cinnamic acid on cell proliferation and apoptosis of nasopharyngeal carcinoma.Methods: Morphology changes of CNE2 cells without treated and treated by CINN were observed with invert microscope.Proliferation changes of CNE2 cells after treated with CINN were detected by MTT colorimetric assay.Flow cytometry was applied to measure apoptosis rates.KLF6,Fas-L,Bcl-2,Bax,P53,cyclinD1 and caspase-3 were measured by Western blot as cell proliferation and apoptosis markers respectively.The interference fragments of KLF6 genes were designed by Designer3.0 software,LV3-shRNA expression vectors were constructed by BamH?and EcoR?double digestion and were transfected into CNE2 cells.The silencing efficacy of KLF6 shRNAs and the changes of apoptosis associated proteins expression were evaluated with QT-PCR and Western blot.Results: After being treated with CINN,CNE2 cells separated gradually marked.The proliferative activity of CNE2 cells treated with CINN were inhibited obviously(P<0.05),showing a dose-dependent and time-dependent apparently.The results of MTT revealed that 2.0mmol/L was the best medical concentration.FCM detected that CINN could induce apoptosis of CNE2 cells.Western blot results showed that after treatment of CINN at different concentrations for 48 h,the expression of KLF6,Fas-L and Bax proteins were significantly increased(P<0.05),the expression of Bcl-2 protein was markedly decreased(P<0.05).After treatment of CINN at the concentration of 2.0mmol/L for different time,the expression of KLF6,Fas-L,Bax,P53 and caspase-3 proteins were significantly increased(P<0.05),the expression of Bcl-2 and cyclinD1 proteins were markedly decreased(P<0.05).After KLF6 gene was knocked down,nearly all of KLF6 expression was blocked and the expression of Fas-L,Bax and TNF proteins were significantly decreased.Conclusions: CINN could inhibit proliferation and induce apoptosis of nasopharyngeal carcinoma cells.The mechanisms might include that,CINN could upregulate the expression of KLF6 and downregulate the expression of cyclinD1 to inhibit proliferation on the one hand,and on the other,CINN could upregulate the expression of KLF6,P53,Fas-L,Bax and caspase-3 and downregulate the expression of Bcl-2 to induce apoptosis.KLF6 might be an upstream gene of Fas-L,Bax and TNF,but further research was needed to uncover whether KLF6 was a key target for CINN inhibiting proliferation and inducing apoptosis of nasopharyngeal carcinoma cells.
