Objective: To investigate the expression and correlation of MIP-1? and S clerostin in multiple myeloma patients and myeloma cell lines.Method:1.A total of 54 MBD inpatients were included in Affiliated Hospital of Gu ilin Medical University Hematology Department from September 2013 to J une 2015.According to the Zhang Zhinan's diagnosis and curative effect standard of blood disease,all subjects were divided into 53 cases group.We chose 53 cases of MBD patients and 30 cases of healthy people and two human myeloma cell lins-RPMI8226,H929 as experimental subjects.Fresh bone marrow of all subjects were collected.At the same,we record ed some laboratory data: microglobulin-2beta,and bone damage level and other biochemical indicators,using ELISA and RT-PCR to evaluation bone marrow supernatant serum and the supematants of cell culture sclerostin and MIP-1? level.2.In vitro experiment,by using specific anti-MIP-1? a nd anti-sclerostin antibodyto neutralize MIP-1? and sclerostin,which secret ed from RPMI8226 and H929 myeloma cells,we set the group which add antibody asthe experimental group,the group whcih do not add the an tibody as the control group:(1)RPMI8226 control group;(2)RPMI8226+MIP-1? antibody intervention group;(3)H929 control group;(4)RPMI8226+sclerostin Antibody intervention group;(5)H929+MIP-1? Antibody interve ntion group;(6)H929+MIP-1? antibody intervention group.ELISA and RTPCR wereused to detect the expression of MIP-1? and sclerostinbetween t he subjects.Results:1.Among the patients and healthy control subjects,the levels of MIP-1? and sclerostin in MM patients bone marrow were obvi ously highe than those in the control group,there was statistically signific ant difference between these two groups(p<0.01).In MBD patients,the MI P-1? and sclerosis levels in C group were significantly higher than those in A groupand B group,there was also statistically significant difference b etweenthese two groups(p<0.05);sclerosis levels were positively correlated with MIP-1?,?2-microglobulin,a Ca and was negativily correlated with ALP,HB,ALB.There was no associated with serum Ca,Plt,Scr,LDH,1,25(OH)D3 and sclerostin level.2.In vitro,both of the two MM cell lin es were able to secrete the cytokines:MIP-1?and sclerostin.But levels of MIP-1? and sclerostin were different betweentwo cell lines.And MIP-1? in two cells was associtaed with sclerostin.By contrasting before and after adding anti MIP-1? and sclerostin antibody,we find that MIP-1? and scle rostin levels was lower than the control groups,therewas statistically signi ficant difference between these two groups(P<0.05);Furthermore,compar ed with the control group,groups which add the anti-MIP-1? antibody the expression of sclerostin also decreased.The same situation also occurred after the addition of anti-sclerostin antibody.The levels of MIP-1? and sc lerostin in experimental groups were positively correlated with the control group.Conclusion: 1.MIP-1? and sclerostin were expressed in most MM patients,and MM cell lines,but the levels in different cell lines was no t he same;2.The level of MIP-1?,sclerostin was positively correlated with tumor load and bone destruction in multiple myeloma;3.In vitro research,we confirm that sclerostin is secreted through the MIP-1? stimulation by myeloma cells,but its mechanism is still unknown.
