Flor-Essence(FE),a herbal tea with eight kinds components of plant,is widely applied for north American cancer patients.A variety of herbal experimental studies have shown that FE has many biological activities including immune stimulation and antitumor effect.We demonstrated recently that FE can alleviate the heavy load movement of rats caused by low immune ability,increase the activity of natural killer(NK)cell to improve immune ability and motor function.Clinical studies have showed that FE can help cancer patients to rebuild immunity and repair the damage of chemoradiation,and thus to improve the quality of life.But it is still not clear about the mechanism of FE resistance to radiation and chemotherapy side effects.Cyclophosphamide(CP),a cytotoxic chemotherapy drug,is the most common clinical treatment of malignant tumor.It can kill tumor cells in non-special manner and cause side effects on the cellular and humoral immune suppression.through influencing cell cycle.Therefore,CP is often used as a drug with low immune mice model in clinical research.In this study,low immune mice model was established with CP induction.Immune organs(spleen and thymus)index,spleen cell proliferation and the expression of PCNA in normal and low immune mice were detected by MTT and Western blot.The effects of FE on immune cells activity,was analyzed by the neutral red method and LDH method.Furthermore,cytokines(INF-?,INF-?,IFN-?,IL-12)expressions in immune organs and peripheral blood were detected by RT-PCR and ELISA to verify the FE effects on immune system function.Finally,to explore the mechanism of FE strengthen immune system function,the expression of signaling pathway related protein Cav-1,NF-?B was investigated by Western blot.The result as follows:(1)The immune organ weight index of normal mice was significantly increased after taking FE,especially,the growth of lymphocytes and the expression of PCNA in spleen by high dose of FE.The phagocytosis viability and NK cell killing destruction ability in normal mice were enhanced by FE.Secretion of IL-12 and IFN-? in serum were increased(P<0.01).The results revealed that FE can significantly enhance the immune indexes in normal mice.The progress was regulated by Cav-1/NF-?B signaling pathway.(2)The low immunity mice model was established with cyclophosphamide intraperitoneal injection.We tested the number of blood cells,spleen index and the secretion of the serum levels of IL-2 and TNF-? and so on.The results showed that the indexes of CP mice declined significantly(P<0.01),which suggested that the mice model of cyclophosphamide immunosuppression had been set up successfully.(3)Splenocytes activity,cytokine secretion in serum,the phagocytosis of macrophage cell and the toxicity of natural killer cells were significantly increased in cyclophosphamide immunosuppression mice model by taking FE(P<0.01).Conclusion: FE promoted the immune function of normal mice and also significantly improved immunosuppression mice caused by cyclophosphamide resistance to radiation and chemotherapy side effects.FE regulated the body's immune system defensing and monitoring function and reduced the side effects of radiation and chemotherapy.These results were important in the clinical treatment with FE.
