| OBJECTIVE Dihydroquercetin is a kind of flavonoids.According to the preliminary study,it has been found dihydrogenquercetin known as strong antioxidant,has very high potential.This study to explore the hepatoprotective effects of dihydroquercetin on acute liver steatosis in mice fed high fat diet plus alcohol binge,through high fat feeding plus acute ethanol binge model.METHODS The mice were fed with high fat diet plus ethanol binge to induce liver steatosis.Serum aspertate aminotransferase(AST)and third transaminase(ALT)levels and TG level was detected after treated dihydrogenquercetin.Also we performed HE,oil red O stain and immunohistology assay of sterol regulatory element-binding protein-1(srebp1).RESULTS:There is no significant difference(P>0.05)in the levels of AST,AIT and TG in serum and hepatic TG between control diet feeding group and normal group.High fat diet feeding group compared with normal group the levels of AST,ALT and TG in serum and hepatic TG increased significantly.High fat diet feeding plus acute ethanol binge group compared with high fat diet feeding group the above index increased more obviously.High,medium and low dose dihydreoquercetin and high fat diet feeding plus acute ethanol binge groups compared with High fat diet feeding plus acute ethanol binge group the levels of AST,ALT and TG in serum and hepatic TG have declined.In addition,did HE,oil red and SREBP-1 immunohistochemical staining.Found that Control diet feeding group and normal group have no obvious changes.But High fat diet feeding group compared with normal group have obvious hepatic steatosis.High fat diet feeding plus acute ethanol binge group compared with high fat diet feeding group the above index increased more obviously.High,medium and low dose dihydreoquercetin and high fat diet feeding plus acute ethanol binge groups compared with High fat diet feeding plus acute ethanol binge group hepatic steatosis has improved and the improvement of the hepatic steatosis more obvious in higher dihydreoquercetin and high fat diet feeding plus acute ethanol binge groups.CONCLUSION Our current research proved high fat diet feeding can form slightly hepatic steatosis,hepatic steatosis caused by high fat diet feeding plus acute ethanol binge group is more obvious.The dihydroquercetin can reduce the liver steatosis in mice fed with high fat diet plus alcohol binge which have protective effect on liver. |
