OBJECTIVES:To evaluate the association of TAS2R38 and TAS2R13 polymorphisms with persistent allergic rhinitis(PER)due to house dust mite in a Chinese Han population.METHODS:In a hospital-based case-control study of 570 patients with mite-sensitized PER and 644 healthy controls.Blood was drawn for serum total IgE and allergic-specific IgE(against Dermatophagoides pteronyssinus and Dermatophagoides farina)assay,as well as genomic DNA extraction.RESULTS:The genotype and allele frequencies of the rs1726866 fromTAS2R38,rs1015443 from TAS2R13 show no significant association with PER induced by mites in general.However,after stratification analyses the CTand CT/TT genotypes of rs1726866 exhibited a significantly decreased risk of mite-sensitized PER(CT:adjusted OR=0.642,95%CI=0.451-0.915;CT/TT:adjusted OR=0.683,95%CI=0.490-0.952)in the subgroup of>18 years old.Moreover,the homozygote TT genotype of rs1015443 exhibited a significantly increased risk of mite-sensitized PER(TT:adjusted OR=1,946,95%CI=0.490-0.952)in the subgroup of male.However,none of these SNPs was correlated with serum total IgE levels and the asthma.The multifactor dimensionality reduction(MDR)software show no significant associations were detected between the 2 selected SNPs.CONCLUSION:Our results suggest that age and gender may have an impact on the association of two SNPs(rs1726866 and rs1015443)with the risk of mite-sensitized PER in a Chinese Han population.Further study on signaling pathways would help in understanding of the pathogenesis of allergic rhinitis and identification of targets for new therapeutic intervention.
