Study On The Secretion Of CAP1 And The Functions Of Secreted CAP1

CAP1,which can regulate PKA kinase system,plays an important role in the cAMP pathway in higher organisms.What's more,CAP1 can regulate the cytoskeleton movement.Recently,Study on CAP1 mainly focus on the relationship between the CAP1 and the tumor cells.The over expression of CAP1 is positively correlated with various types of tumors,but the regulation mechanism is not clear.Further research on the function of CAP1 protein and its relationship with tumor cell migration will provide a target for tumor therapy.We found that CAP1 can secret into the culture medium in the case of some stress,such as serum starvation and glucose starvation.The secretion of CAP1 can be detected in 1 Hours under stress of serum starvation.We added the purified proteins into the culture medium and found that most of the CAP1 adhered to the surface of the cell membrane,and a small portion of CAP1 was able to penetrate the cell membrane into the interior of the cells.Then,we have studied the functions of CAP1.Addition of purified CAP1 proteins into the culture medium could significantly stimulate the GLUT1/GLUT4's movement from Vesicles to membrane and increase the absorption and utilization of glucose.Through cell wound healing and Transwell experiments,we found that the cells cultured in the medium with purified CAP1 had a significant increase in cell migration.Thus the purified CAP1 plays an important role in cell migrating.The overexpression of CAP1 has been reported repeatedly in various tumor tissues.While we found that the CAP1 protein is a secreted protein.The overexpressed CAP1 can secret into the microenvironment of tumor cells,which will increase the content of CAP1.As a result,the growth and migration of tumor cells have been promoted.Our study provides a possible target for the treatment of cancer.By injecting human CAP1 antibodies to cancer patients,the CAP1 protein content in the tumor microenvironment should be reduced,50 as to inhibit tumor growth and metastasis.Absolutely,furthher studies should be carried out to verify this assumption.
