Rhamnus utilis,which belongs to Rhamnacease Rhamnus,is a famous dyestuff plant.Known as "Chinese green",its pigment for dyeing enjoys high international reputation in aspects of printing and dyeing plants in history.In addition to being natural green dye pigments,Rhamnus utilis is widely used as anti-inflammatory herb in provinces around the Yangtze River,so our group carried out the study on anti-inflammatory and analgesic activity.Results are as follows:(1)The optimization of extracting Rhamnus utilis seed oil was studied by using uniform design,and the optimal extraction conditions was as follows:the ratio of material and liquid 1:19(g:mL),extraction temperature 90 ?,extraction time 2 h.Under these conditions,theoretical predictions of seed oil yield was 22.14%and the actually average yield of oil was 22.47%.By GC-MS,a total of 21 fatty acids were identified in the oil.The relative content of linoleic acid was 39.54%in the oil,followed by oleic acid(30.14%)and linolenic acid(21.53%),totally accounted for 91.21%.(2)After studying the extraction process and the physicochemical properties of Rhamnus utilis fruit pigment,optimum extraction conditions were as follows:acidified ethanol 70%,extraction temperature 90 ?,ratio of material and liquid 1:12(g:mL),extraction time 2.5 h.The physicochemical properties study showed that the pigment had a development value,which provided a reference for the development and utilization of Rhamnus utilis fruit pigment.(3)The study on acute toxicity with mice showed that Rhamnus utilis fruit had a certain toxicity and was not edible.(4)In the hot-plate tests with mice,different dose groups of Rhamnus utilis leaf butanol extracts all exhibited significant inhibition(P<0.01).The pain threshold increase rate of high-dose group reached 63.48%,which was very close to the positive control drug aspirin group.(5)In the writhing experiments,Rhamnus utilis leaf butanol extracts were also exhibited significant inhibition(P<0.01).Compared with model group,writhing latency of high-dose was extended to double time,and the rate of inhibition was 62.16%,which was close to the aspirin group.(6)In the xylene-induced ear edema experiments,the results showed that Rhamnus utilis bark and leaf had significant inhibition(P<0.05,P<0.01).Both of them showed outstanding anti-inflammatory activities,and the influence on anti-inflammatory of leaf was better than that of bark.(5)The anti-inflammatory of eluted fractions,which were obtained from Rhamnus utilis leaf by silica gel column chromatography,were studied.The results showed that high-dose group and low-dose group of the ratio of chloroform and methanol 9:1 fraction had significant inhibitionon ear swelling in mice(P<0.01).The swelling inhibition rates were 63.47%and 50.98%respectively,and the high-dose group was very close to positive control group(67.13%).(6)F lavono ids and saponins was contained in the fractions under the ratio of chloroform and methanol 9:1 and 7:3 from Rhamnus utilis leaf butanol extracts by silica gel column chromatography.Basically,there was no flavonoids and saponins in other fractions.Two compounds were obtained under the ratio of chloroform and methanol 1:9 by semi-preparative HPLC separation.
