The Study Of Tricin On Purification And Anti-allergic Activity

Objective We obtained tricin from bamboo leaves flavonoid through isolation and purification.On the base of it,we explored how did tricin make effects on experiments such as reaction of allergen and other lists about anti-anaphylaxis trials in vivo and in vitro,which that provide evidence on the study of tricin on anti-allergic activity.Methods Silica gel column chromatography and polyamide column chromatography are used to get pure tricin from bamboo leaves flavonoids.Thin-layer chromatography and polyamide plate are used to track the ingredient of tricin.High performance liquid chromatography-evaporative light scattering device are adopted in the determination of tricin.By using area normalization method,the content of tricin is calculated.In the experiment of smooth muscle tension from guinea pig which mainly adopted sensitized guinea pig and normal ones.In the whole experiment,we designed rat skull periosteum mast cell degranulation test and DNFB-induced mouse type ? hypersensitivity(delayed-type hypersensitivity;DTH)test.Results Through determination,we measured that the percentage of tricin is 98.09%.The outcome demonstrated that tricin final concentration of 1×10-5,2×10-5 and 4×10-5 g/ml,could significantly antagonize the increase of ileum smooth muscle tension caused by the OVA and inhibited that histamine augmented the tension of ileum contraction in normal guinea pig.We found that,compared with the vehicle control,different dose of Tricin can inhibit the degranulation of mast cells,tricin concentration can also reduce the swelling degree of DTH response in mice,cut down mouse thymus index and spleen index.Conclusion It initially hinted that tricin may be have the effect of anti-allergic.Probablely by stabilizing mast cell membrane,inhibition of mast cell degranulation,inhibition of allergic reactions to achieve.
