| Objective;Based on the previous study in the laboratory about the significantly elevating peak area of an endogenous physiological substance which was induced by Sinisource.This present research study aimed to achieve and to determining its structure.Methods:Sinisource lyophilized powder was induced into rat for analyzing the cerebrospinal fluid;endogenous substance and for screening to make physiological characteristics.Ten SD rats were randomly divided into two groups,5 each.First group were received 4.9 g/kg freeze dried Sinisource,and another group were received water which served as blank orally by gavage technique once a day for seven days.Five hours after the last administration,the cerebrospinal fluid of rats from each groups were extracted and were separately collected via foramen magnum.The significantly change of endogenous physiological substance from cerebrospinal fluid were determined and analyzed.The physiological structure identification was done using cerebrospinal fluid from aforementioned experiment.The structure identification was screened for endogenous physiological substance using UPLC/Q-TOF-MSResults:1.In the view of endogenous physiological substance feature marker from rat’s cerebrospinal fluid analysis,Sinisource lyophilized powder was induced endogenous physiological substance in the experimental animal.The results revealed that the cerebrospinal fluid from treatment group had the same peak number of endogenous physiological substance but different in the peak area size.2.In view of cerebrospinal fluid endogenous physiological substance analysis and characteristic marker screening using UPLC/Q-TOF-MS,the retention time of the main ingredient in rat cerebrospinal fluid of the blank control group and the treatment group(tR)were compared using 12 Mass Spectrometry positive ion mode.The results showed that the endogenous physiologically substance in cerebrospinal fluid from treatment group peak area at a retention time of 8.59 min statistically showed higher than 6.90 min.(p<0.05)3.In view of Sinisource lyophilized powder inducing the rat cerebr-ospinal fluid endogenous physiological substance structure identification,the results exhibited that the formula of endogenous physiological subs-tance from rat’s cerebrospinal fluid which was induced by Sinisource 1-yophilized powder is C16135NO2.In conclusion:1.Sinisource powder did not induce the changing of endogenous ph-ysiological substance in rat cerebrospinal fluid,but change the size of the peak area of an endogenous physiological substance.2.The formula of endogenous physiological substance from the rat’s cerebrospinal fluid which was induced by Sinisource powder is C16H35NO2;furthermore,it was initially speculated as Sphingosine substance. |
