High-salt diet,which can lead to high blood pressure and other cardiovascular disease,is an important external threat for human health.The rise of serum homocysteine(Hcy)level will cause Hyperhomocysteinemia(Hhcy).Studies have shown that Hhcy is an independent risk factor for cardiovascular disease.High-salt diet and Hhcy are both associated with cardiovascular disease,but there is little research on the relationship between the high-salt diet and Hhcy.In the paper,the effects of high salt stress on the metabolism of Hcy on animal and cellular level were studied.Betaine and taurine,which are osmotic pressure regulators and related to Hcy metabolism,were selected to determine whether they were antagonistic to high salt stress on Hcy metabolism.This work provide theoretical guidance for further study the relationship between high-salt diet and Hhcy and Hhcy prevention and treatment.On the cellular level,cells were stressed with different concentrations of NaCl,and betaine intervention,taurine intervention,and contervention of betaine and taurine were imposed.The expression of cystathionine β-synthase(CBS)and betaine-homocysteine S-methyltransferase(BHMT),two key enzymes of Hcy metabolism,were detected by Western blot.The effect of high salt stress on Hcy metabolism,and the function of two osmotic pressure regulators in proteomics were studied on protein level.On animal level,mouse model was developed throngh excessive intake of high-salt diet,and intervened by betaine and taurine intervation were gaven by free water drinking.Level of MDA content,SOD vitality,expression of CBS and BHMT in liver and kidney,and levels of serum osmolality,Hcy,blood sugar and other indicators were compared.The relationship between stress and high salt Hcy metabolism were studied on gene transcription level.It was showed that high salt stress inhibited expression of CBS and BHMT in cells,and betaine intervention could effectively antagonize the high salt stress on CBS and BHMT inhibition.High-salt diet resulted the lower weight gain,higher liver and kidney ratio,damaged antioxidant system,and elevated levels of blood glucose,serum urea,serum osmolality and Hcy,and inhibited expression of CBS and BHMT.Betane effectively ameliorate the blood glucose,serum urea,serum osmolality,and Hcy levels,and the expression of CBS and BHMT.Taurine effectively ameliorate the damage of antioxidant system.In summary,high salt stress inhibited the expression of CBS and BHMT,and led to elevated Hcy level,through increasing osmotic pressure.Betaine had significant therapeutic effect,but the therapeutic effect of taurine needs further researches.
