Objective:To elucidate the changes of betatrophin in the triglyceride metabolism,one of the cytokines secreted by the liver,and the intervention effects of FTZ on it after the thesis "the liver is the pivot to regulate the lipid and get rid of the waste" to explore the dynamic change and mechanisms.Methods:We collected the blood samples of high triglycerides patients,type 2 diabetes patients,high triglycerides patients with type 2 diabetes and healthy people and determined the amounts of betatrophin in it using ELISA assay.C57BL/6j male mice were fed on diets with 10%fat content(MD10%fat),45%fat content(MD45%)to establish hyper-triglycerides animal model,then we monitored each group the content changes of glucose,total cholesterol and triglyceride after sixth weeks and oral glucose tolerance test(OGTT)were given at 12th weeks,16th weeks respectively to observe the sugar and fat tolerance changes.The effects of FTZ on blood lipid,blood glucose level in mice were also observed.Tolerance test were given at the end of the sixteenth week,nineteenth weeks respectively.Oral glucose tolerance test(OGTT),oral fat tolerance test(OFTT)were used in mice in vivo to observe the metabolism of glucose and lipid.The tissue homogenates of important organs(liver and muscle)were used to detect glycolipid metabolism changes within the lipid content.Reverse transcription polymerase chain reaction(RT-PCR)technique was used to detect the expression quantity of of betatrophin expression in the liver of mice under different states and other related gene for lipid metabolism changes in liver.Results:The plasma betatrophin content of high TG patients and high TG compared with high FPG patients showed a downward trend compared to the normal people,which has no statistically significance,however,betatrophin contents of only high TG patients decreased more obvious than that merged with high FPG.The plasma betatrophin contents of type 2 diabetes patients exhibited slightly higher than that of normal,still without statistically significance.Compared with the normal group,the high fat diet can.induce hyper-triglycerides model in C57BL/6J mice.GLU,TC,TG content of the model group were significantly increased,and the increase of body weight of model mice was significantly greater than that of normal group,a significant increase in body weight and viscera visceral adipose tissue compared with those in the model group;the content of liver lipids(triglyceride)increased significantly,and that in the muscle tended to increase.Second,compared with the model group,FTZ made the GLU,TC,TG content decrease to some extent,the lipid content of liver and muscle decreased significantly(triglyceride).Reverse transcription polymerase chain reaction(RT-PCR)detection results showed SCD1,Gm6484,Acc,Fas expression decreasing in the liver,while increasig the expression of LPL.Conclusion:The clinical results suggested that hyper-riglycerides and hyperglycemia both have the effect on betatrophin secretion,indicating that the betatrophin gene expression may be regulated by hyper-triglycerides and hyperglycemia;what' s more,the two factors should have a synergistic effect.The clinic regulating of the gene in hyperglycemia and hyper-triglycerides patients still needs further research to elucidatde the details and mechanisms.High fat purified diet successfully replicated the mouse model of hyper-triglycirides.The model has the characteristics of high blood glucose and high triglycerides and FTZ reduced the plasma triglycerides amounts and iver,muscle tissue deposition of triglycerides.It is suggested the expression of betatrophin in the liver regulate the plasma triglyceride metabolism.After the administration of FTZ,the expression of the gene betatrophin decreased in the liver glucose and lipid metabolism related genes(SCD1,Acc,Fas)expressions decreased in the liver.Those...
