Objective: To study the chemical constituents in the fruit of Swietenia macrophylla with degreasing alcohol extracts which posseses hypoglycemic activity,and search the lead compound with hypoglycemic activity in the degreasing alcohol extracts by isolating and purifying its chemical constituents using new techniques.Method: After soaked in petroleum ether,the ester compounds of the extraction was analyzed by GC-MS,and the residue was percolated in 95% ethyl alcohol till the leaching solution getting colourless.Then volatilized ethyl alcohol and extracted by ethyl acetate.The constituents was isolated and purified by column chromatography on silica gel,RP-18,Sephadex LH-20,and HPLC,and identified by physicochemical properties and spectroscopic method including 1D and 2D NMR,MS,UV,IR,et al.Four alloxan appliacation to the model mice,metformin hydrochloride as control drug,investigation to the the fruit of Swietenia macrophylla aqueous extract of lowering blood glucose and glucose tolerance levels.Result: 16 volatile constituents were identified via GC-MS analysis from the part of petroleum ether of the fruit of Swietenia macrophylla.These constituents,mainly Fatty acid and its ester,account for 99.70 % of the total content.9,12-octadecadienoic acid(27.73%),8-octadecenoic acid(28.67 %),9,12,15-octadecatrienoic acid(13.45 %),octadecanoic acid(12.77 %)are highly-contained in this part,where the content of fat is above 55 %.21 compounds were isolated and identified as swieteliacate A(1),swieteliacate B(2),swieteliacate C(3)and swieteliacate D(4),swietenine(5),swietenine acetate(6),febrifugin(7),khayasin T(8),3-O-Tigloyl-6-O-acetylswietenolide(9),3-O-tigloyl-swietenolide(10),3,6-O,O-diacetylswietenolide(11),fissinolide(12),3-O-acetylswietenolide(13),proceranolide(14),swietenolide(15),swietemahonin E(16),swietemahonin F(17),7-deacetoxy-7-oxogedunin(18),secomahoganin(19),altissimanin B(20),odoratone(21).The extracts of Swietenia macrophylla displayed the activities of adjusting theglucose tolerance and hypoglycemic activity against diabetes induced by alloxan on mice.Moreover the role of Swietenia macrophylla presents certain dose-effect relationship,though not significant.The glucose tolerance of normal mice also had a certain adjustment function..Conclusion: These constituents,mainly fatty acid and its ester;and compounds1-4 are new compounds and compounds 7,9,12-14,20,21 were found from Swietenia macrophylla for the first time in the ethyl acetate part.The role of Swietenia macrophylla presents certain dose-effect relationship,though not significant.The glucose tolerance of normal mice also had a certain adjustment function..
