| Objective:To investigate the level of CD4~+CD25~+FOXP3~+T and CD8~+CD25~+FOXP3~+T cells in peripheral blood of patients with ankylosing spondylitis(AS),and to investigate the effects of low-dose recombinant human IL-2.Methods:In the first part,We collected 73 newly diagnosed patients with AS in our hospital from March 2017 to September 2017,including 60 males and 13 females,aged 18-74,with an average age of 38.70±14.23 years.All the patients met the diagnostic criteria for AS revised in New York City in 1984,voluntarily signed the informed consent form and the Bath AS Disease Activity Index(BASDAI)form.Meanwhile a total of 85health persons were selected as controls,including 70 males and 15 females,aged21-65,with an average age of 39.63±13.71 years.There is no significant difference in the basic data such as gender and age between the two groups.The two groups of subjects were taken venous blood which was sent to our laboratory laboratory in the morning,and detect CD4~+CD25~+FOXP3~+Treg(CD4~+Treg),Th17,Th17/CD4~+Treg,CD4~-CD25~+FOXP3~+(viewed as CD8~+Treg),CD4~+/CD8~+Treg,comparing the test results of AS group with that of healthy control group.In the second part,we collected the erythrocyte sedimentation rate(ESR)and c-reactive protein(CRP)of the AS group,analysising the correlation of CD4~+Treg,Th17,Th17/CD4~+Treg,CD8~+Treg,CD4~+/CD8~+Treg levels with ESR,CRP and BASDAI scores of AS group.In the third part,we selected 48 AS patients as IL-2 treatment group(50WIU,subcutaneous injection,continuous use for 5 days),analysising of the CD4~+Treg,Th17,Th17/CD4~+Treg,CD8~+Treg,CD4~+/CD8~+Treg levels before and after treatment with IL-2.Results:Compared with healthy controls,the counts of CD4~+Treg and CD8~+Treg in peripheral blood were lower in AS patients than in healthy controls(P=0.043,P<0.001).The Th17 counts,Th17/CD4~+Treg,and CD4~+/CD8~+Treg were higher than those in healthy controls(P=0.012,P=0.018,P<0.001).There was no correlation between CD4~+Treg,Th17,Th17/CD4~+Treg,and CD4~+/CD8~+Treg levels in AS group with ESR and CRP.The CD8~+Treg,Th17,CD8~+Treg,CD4~+/CD8~+Treg levels and BASDAI analysis were no correlation,the CD4+Treg,Th17/Treg levels were correlated with BASDAI.The CD4+Treg,Th17,CD8+Treg,CD4+/CD8+Tregs were higher after treatmentwithIL-2(P<0.000,P<0.000,P=0.001,P<0.000),the Th17/CD4~+Treg was decreased after treatment with IL-2(P<0.000).Conclusion:1.The counts of CD4~+Treg and CD8~+Treg in peripheral blood of AS patients are decreased,the Th17 counts and Th17/CD4~+Treg are increased.2.IL-2 have the effect of increase the levels of CD4~+Treg,Th17,and CD8~+Treg cells,the ability of IL-2 to promote CD4~+Treg cells was stronger than that of Th17.3.There is a correlation between CD4~+Treg,Th17/CD4~+Treg and BASDAI scores.There was a negative correlation between CD4~+Treg and BASDAI score,but a positive correlation between Th17/CD4~+Treg and BASDAI score. |
