Objective:To observe the changes of the cell morphology of cardiac tissue and cardiovascular endothelial cells under CIH conditions;To investigate the expression of PHD3 in cardiovascular tissues of CIH rats and analyze sis its correlation with CIH.Methods:Twenty-four male adult healthy SD rats were selected.They were randomly divided into three groups according to the intermittent hypoxia duration time:normal control group,4-week CIH group and 6-W CIH group.At the 4th and 6th week when the CIH model group was established,half of the rats were randomly selected from each group?Then three groups of rat heart tissue specimens were taken and embedded routine paraffin.Immunohistochemical procedure:after routine paraffin sections and dewaxing;then through antigen retrieval,the addition of primary antibody ang secondary antibody,DAB coloration,counterstaining,differentiation,transparence,and mounting,etc.;followed by immunohistochemistry;finally getting conclusion?Statistical Analysis:SPSS 24 statistical software was used to analyze the data;One-way analysis of variance was performed;LSD-t test was used to perform multiple comparisons between groups.Mean±SD(~_x±s)count data;correlation analysis use Pearson correlation.Results:(1)Cardiac HE staining:1 normal control group,myocardial cells and fibers are arranged more regularly,no myocardial cell swelling;24 weeks CIH group,myocardial cells and fibers are arranged more regularly,myocardial cells are mild edema,intravascular expansion,hyperemia,and small blood vessel formation Increased,a few inflammatory cells;36 weeks CIH group,myocardial cell and fiber arrangement disorder,fracture,myocardial cell edema,vasodilatation,congestion,increased tubule formation,more inflammatory cells.(2)Cardiac vascular tissue PHD3 expression:1 compared with the normal group,PHD3 positive expression rate of 4 weeks CIH group decreased(t=5.041,P<0.001),the difference was statistically significant;2 compared with the normal group,6 weeks The positive expression rate of PHD3 in the CIH group was significantly decreased(t=8.587,P<0.001),and the difference was statistically significant.Compared with the CIH group at 4 weeks,the positive rate of PHD3 in the 6-week CIH group was decreased(t=2.899,P).<0.05),the difference was statistically significant.Conclusion:(1)CIH can cause heart tissue damage and is related to the cycle time of CIH;(2)CIH can cause the decrease of PHD3 expression in cardiac vascular endothelial cells,and the decrease of PHD3 is related to the prolongation of CIH.
