| Objectives:The effect of this wound on DFU healing in rats was studied by means of self-made gelatin/bletilla glue-notoginseng porous materials and rat diabetic foot ulcer model,and the mechanism of promoting ulcer healing was discussed.Methods:1.Using the ball mill method to prepare the ultrafine rice level notoginseng,apply the freeze-drying method to prepare gelatine/bletilla glue-notoginseng porous materials and gelatin/white and rubber porous materials;2.SPF(Specific Pathogen Free)health level 60 SD male rats,only four weeks,high-sugar high-fat feeding weighing of rat,according to random number table method is divided into material matrix,gelatin sponge group,the control group and vaseline control group,blank(sterile gauze),each group of 15.Use shaving shaving machine,in the back on both sides of the spine with a magnet to make 1 cm diameter size of animal model of rats back full-thickness skin defect with gelatin/bletilla glue-notoginseng porous materials,gelatin sponge,vaseline gauze material,sterile gauze(soaked in water for injection)covering the wound(with sterile dressing and a bandage fixation,dressing change once a day),single cage to prevent lick each other;Respectively in 3d,7d after injury(each 5 only)in each group,with H.E staining to observe the wound healing of constituting the testing groups with Western Blot interventions to wound tissue beta serial element(?-catenin),glycogen synthesis kinase-3?(GSK-3?),R-spine protein 3(Rspo-3)expression.Results:1.Each team will then observe the wound healing of the,wound healing rate in 3d,7d had significant difference(p < 0.05),the material base of capillary growth in the wound in the other group is the best;2.There were differences in the positive expressions of each group at various points of time(p<0.05),and the positive expression of ?-catenin and rspo-3 was the highest in the granulation tissue of the material treatment group,and the lowest expression was gsk-3?.Conclusion:Gelatine/bletilla glue-notoginseng porous materials are safe new wound dressings;In the rat model of DFU its can obviously shorten the wound healing time,speed up the wound repair,promote wound healing mechanism may be related to beta serial element(?-catenin),glycogen synthesis kinase-3?(GSK-3?)low expression,R-spine protein 3(Rspo-3)of high protein expression level. |
