P2X7 Receptor Involved In Contralateral And Ipsilateral Moxibustion-induced Analgesia

Objective:To compare the analgesic effects between contralateral and ipsilateral moxibustion and the relation with P2X7 receptor in C57BL/6J mice.Methods:All mice 20±2g were used in this experiments and were randomly divided into control group,model group(CFA group).Injected with complete Freund's adjuvant(20ul sigma)to the left hind paw for inducing inflammatory pain.Four days later,the model group were randomly divided into ipsilateral ipsilateral moxibustion group,contralateral moxibustion group,ipsilateral A438079 group,contralateral A438079 group,ipsilateral moxibustion+ipsilateral A438079 group,ipsilateral moxibustion+contralateral A438079 group,contralateral moxibustion+ipsilateral A438079 group,contralateral moxibustion+contralateral A438079 group.Ipsilateral moxibustion group and contralateral moxibustion group were given moxibustion for 30min;others were given A438079 injection and moxibustion accordingly.Pain thresohold test was tested before,after the intervention.Statistical analysis was performed with the SPSS 19.0 and figures were made by GraphPad Prism 6.0.Results:1.The analgesic effects of moxibustionCompared with the control group,the pain threshold of CFA group was great decreased after CFA injection(P<0.01);compared with the CFA group,the pain thresholds of both ipsilateral and contralateral moxibustion groups were increased after intervention.There were statistical difference in ipsilateral moxibustion group after intervention at 30 min,60min,90min(P<0.05).There were statistical difference in contralateral moxibustion group after intervention at 30 min,60min(P<0.05).2.Comparison in analgesic effects between ipsilateral and contrateral moxibustionPain threshold increased in both ipsilateral and contralateral moxibustion.There was no statistical difference between ipsilateral and contralateral moxibustion(P>0.05).3.Comparison analgesic effects between ipsilateral and contralateral A438079 involvementCompared with the control group,the thresholds decreased in ipsilateral and contralateral A438079 group(P<0.01).Compared with CFA group,the pain threshold of ipsilateral and contralateral A438079 group was increased(P<0.05).Compared with CFA group,the threshold of ipsilateral A438079 group were increased and sustained at 90 min.There was statistical significant in contralateral A438079 group at 90 min,compared with CFA group(P<0.05).Compared between ipsilateral and contralateral A438079 group,there were statistical significant at 30 min and 60min(P<0.05).4.Comparison between ipsilateral and contralateral moxibustion withi A438079 involvement(1)Compared with CFA group,there was no statistical significant in ipsilateral moxibustion+ipsilateral A438079 group(P>0.05);there was statistical significant in ipsilateral moxibustion+contralateral A438079 group at 30min(P<0.05),but no statistical significant at 60 min,90min,120 min and 150min(P>0.05).(2)Compared with CFA group,there was statistical significant in contralateral moxibustion+contralateral A438079 group(P<0.05);there was no statistical significant in contralateral moxibustion+ipsilateral A438079 group(P>0.05).Conclusions:1.Moxibustion has analgesic effect in inflammatory pain;2.The analgesic effects were similar between ipsilateral and contralateral moxibustion;3.The P2X3 receptor was involved in the analgesic effects of moxibustion:(1)Analgesic effects in ipsilateral and contralateral moxibustion were inhibited within ipsilateral A438079 injection;(2)Analgesic effects of ipsilateral moxibustion decreased within contralateral A438079 injection;the analgesic effects increased in contralateral moxibustion within contralateral A438079 injection.
