Effect Of Acupuncture On Expression Of Brain Gut Peptide Motilin In Anxiety Model Rats

Purpose The study was aimed to investigate the effects of acupuncture on the contents of motilin and its receptor in both central amygdala and duodenum by stimulating "Guan yuan" and "Xiajuxu" acupoints on chronic unpredictable stress rats to explore the effect of MTL and its receptors in brain and intestine on the anti anxiety effect of acupuncture on regulating Fu organs.Methods Healthy and male SD rats were randomly divided into 3 groups,blank group,model group and acupuncture group(n=12)by using SPSS21.0 statistical analysis software according to random principle after being domesticated adaptively for 1 week.Rats in both model and acupuncture group were treated with CUS methods to establish the model of anxiety,continuously modelled for 15 days,and acupuncture group's rats were needled Guanyuan and Xiajuxu by mild reinforcing and attenuating once every other day since modeling,Xiajuxu around the turn,1 times a day,30 minutes/day until the end of the modeling,and acupuncture treatments were operated for 8 times in total.The behavioral effects were observed by the elevated plus maze test.The contents of motilin and its receptor in both amygdala and duodenum and corticosterone in the plasma were detected by enzyme linked immunosorbent assay.Results Behavioral results:compared with the blank group,the OT% of model group was decreased significantly(P<0.05),and the OE% was on the decline(P=0.051);besides,compared with the model group,the OT% of acupuncture group was increased significantly(P<0.05),and the OE% value was uptrend(P=0.054);while compared with the control group,both OT% and OE% of acupuncture group had no significant difference(P>0.05),which were close to the blank group.Results of MTL,MTLR in the amygdala:compared with the control group,MTL of model group was decreased(P=0.067);compared with the model group,MTL concentration of acupuncture group was increased slightly;while MTLR in amygdala of blank,model and acupuncture group was not showed statistical differences(P>0.05).Results of MTL,MTLR in the duodenal:compared with the control group,the concentration of MTL in both model group and acupuncture group were decreased slightly;but no significant difference between the groups was shown(P>0.05),besides,compared with the blank group,the concentration of MTLR in the model group were on the decline(P=0.078),compared with the model group,concentration of MTLR in the acupuncture group was increased slightly,which was close to the blank group,but there was no significant difference among the 3 groups(P>0.05).Results of plasma CORT:compared with the blank group,plasma CORT of model group was increased slightly;compared with the model group,that of acupuncture group was on the decline(P=0.065);compared with the control group,the plasma CORT content of acupuncture group was no significant difference(P>0.05).Conclusions Acupuncture "Guan yuan" and "Xiajuxu" acupoints has a good effect of anti-anxiety.Besides,the effect of acupuncture “regulating Fu organs” on anti anxiety may be related to up-regulating the level of MTL in the amygdala and MTLR in the duodenum of CUS rats and down-regulating the activity of hypothalamic pituitary adrenal axis.
