Objective:We observed the expression of tyrosine kinase receptor(Trk receptor)in the facial nerve and facial nucleus of rabbit model of facial nerve injury by Guan-moxibustion therapy,to explore the correlation between the repair and regeneration of facial nerve injury by Guan-moxibustion therapy and Trk receptor.Methods:1.64 healthy Japanese white rabbits were selected,Male and female each of half,Weight 2 ~ 2.5kg.The experiment was divided into two batches,each batch of experimental animals were taken only 32.One batch was used for RT-PCR detection,and one batch was used for immunohistochemistry and in situ hybridization.The animals were randomly divided into four groups,that is,sham operation group,model group,guan-moxibustion group,hyperthermia group,each group of 8.2.Mechanical compression of facial nerve.Buccal branch of facial nerve of rabbits in the model group,the Guan-moxibustion group and the hyperthermia group were separated and squeezed by a special hemostatic forceps;In the sham operation group,only the buccal branch of the facial nerve was exposed and separated,Do not compress,then suture.3.Intervention means:After five days of anti infection treatment,Sham operation group and model group were not treated;The guan-moxibustion group was treated with guan-moxibustion;The treatment method of the hyperthermia group seem to the guan-moxibustion group,Just to charcoal replace the moxa.once a day,six days for a course of treatment,continuous treatment interval between two courses,for the day.4.Index detection:?Observe the general condition;?The expression of TrkA,TrkB and TrkC mRNA in rabbit facial nerve and facial nucleus was detected by RT-PCR and in situ hybridization(ISH);?The expression of TrkA,TrkB and TrkC protein in rabbit facial nerve and facial nucleus were detected by immunohistochemistry.Results:1.Observe the general condition:There was no facial paralysis in sham operation group;After treatment,The recovery of facial paralysis was basically restored in the Guan-moxibustion group;The performance of facial paralysis was slightly improved in the hyperthermia group;The model group showed no significant improvement in facial paralysis.2.Expression of Trk receptor mRNA : The results of RT-PCR and in situ hybridization showed that the mRNA expression of TrkA,TrkB,TrkC in facial nerve,TrkA and TrkC in the facial nucleus of the sham operation group was higher than that of the model group(P<0.05 or 0.01);Compared with the model group,The expression of these indexes was increased in the guan-moxibustion group(P<0.05 or 0.01),There was no significant difference in the expression of these indexes in the hyperthermia group(P > 0.05).Compared with the model group,there was no significant difference in the mRNA expression of TrkB in the facial nucleus of the guan-moxibustion group and the hyperthermia group(P>0.05).3.Expression of Trk receptor protein : The protein expression of TrkA,TrkB,TrkC in facial nerve and facial nucleus of the sham operation group was higher than that of the model group(P<0.05 or 0.01);Compared with the model group,The expression of these indexes was increased in the guan-moxibustion group(P<0.05 or0.01);The protein expression of TrkB in facial nerve,TrkC in the facial nucleus of the hyperthermia group were increased(P<0.05 or 0.01),There were no significant changes in other indicators(P>0.05).Conclusion:1.Guan-moxibustion therapy can treat facial nerve injury in rabbit model of facial paralysis.2.Guan-moxibustion therapy can promote the expression of Trk receptor in the facial nerve and facial nucleus of facial nerve injury model rabbit.It is suggested that one of the mechanisms involved in the repair of facial nerve injury is related to the expression of Trk receptor in the rabbit facial nerve and facial nucleus.
