Objective:To investigate the correlation between Klotho protein and podocyte associated molecule beta-catenin protein and nephrin protein in patients with idiopathic membranous nephropathy(IMN).Methods:We collected 46 cases renal tissue from patients who were first diagnosed as IMN,and19 cases of normal renal tissues were detected by the expression of Klotho protein and beta-catenin protein through immunohistochemical method.ELISA method was used to detect Klotho and nephrin protein from 30 normal subjects and 46 IMN patients.The correlation between Klotho protein,beta-catenin and the expression of nephrin were analyzed.Results:(1)Difference analysis:through variance analysis,the expression of Klotho and beta-catenin expression compared with the control group were statistically significant(P<0.05),and with the progression of the disease,less expression of Klotho in renal tissue,beta-catenin expression was decreased too;Compared with the control group the levels of Nephrin,Klotho in plasma and urine of IMN patients was statistically significant(P<0.05),and with the progression of the disease,the levels of Klotho was declined,but the levels of Nephrin was increased;(2)Correlation analysis:the expression of Klotho and IMN's classification was negatively correlated(r=-0.939,P<0.05),the expression ofbeta-catenin and IMN's classification was negatively correlated(r=-0.962,P<0.05)and positively correlated with Klotho(r=0.906,P <0.05);Klotho in plasma of IMN patients was positively correlated with Klotho in urine(r=0.427,P<0.05),and was negatively correlated with Nephrin in urine(r=-0.320,P<0.05);Klotho in urine of IMN patients was negatively correlated with Nephrin in urine(r=-0.411,P<0.05);Nephrin in urine was positively correlated of IMN's classification(r=0.592,P<0.05),Klotho in renal tissue was negatively correlated with the 24 h urinary protein quantitative(r=-0.696,P<0.05),and was positively correlated with ALB(r=0.724,P<0.05);beta-catenin in renal tissue was negatively correlated with the 24 h urinary protein positive(r=-0.713,P<0.05)and was positively correlated with ALB(r= 0.780,P<0.05);Klotho in plasma was negatively correlated with the 24 h urinary protein(r=-0.444,P<0.05)and was positively correlated with ALB(r=0.518,P<0.05);Klotho in urine was negatively correlated with urinary 24 h urinary protein(r=-0.466,P<0.05)and was positively correlated with ALB(r=0.427,P<0.05);Nephrin in urine was positively correlated with 24 h urinary protein(r=0.313,P<0.05)and was negatively correlated with ALB(r=-0.371,P<0.05).Conclusion:Podocyte injury is the major factor in IMN patients,the expression of Klotho protein was positively correlated with beta-catenin,and was negatively correlated with Nephrin in urine.Klotho decreased with the aggravation of pathology,prompting that Klotho may play a role in podocyte injury.
