Objective:(1)To evaluate the count of CD8+CD25+FOXP3+T and CD4+CD25+FOXP3+T cells in SLE patients;(2)To discuss correlation between CD8+CD25+FOXP3+T CD4+CD25+FOXP3+T cell level and SLEDAI score,disease course,ESR,CRP,C3,C4 and autoantibody.(3)To explore the effect of low dose interleukin-2 on the level of CD8+CD25+FOXP3+T and CD4+CD25+FOXP3+T cells in SLE patients.Methods: This study included a total of 103 healthy people(normal control group).Eighty two systemic lupus erythematosus(SLE)patients were enrolled.The diagnosis was consistent with the SLE classification criteria revised by the American College of Rheumatology in 1997.Collect and record in detail the general data(age,sex,duration,medication),inflammation markers(ESR,CRP,C3 and C4),SLE activity index(SLEDAI score)and autoantibodies(antibody titer of ANA,anti-ds DNA antibody and anti-Sm antibody)of selected SLE patients.The absolute count of CD8+ Treg cells and the absolute count of CD4+ Treg cells in SLE patients were measured by flow cytometry.Thirty of these patients were initially treated(without medical treatment at the time of admission).Compare the levels of CD4+Treg and CD8+Treg cells in normal controls and initially treated SLE patients.And analyze its correlation with disease course,SLEDAI score,ESR,CRP,C3,C4 and autoantibodies.Seventy six patients were treated with IL-2(50 WIU/day×5 days).The absolute count of CD8+Treg cells and the absolute count of CD4+ Treg cells in SLE patients before and after treatment of low-dose IL-2 were measured by flow cytometry.All the experimental data were analyzed by SPSS 22.0 software.Results:(1)The number of CD4+Treg cells in SLE patients was lower than that in the normal control group(P<0.001);the number of CD8+Treg cells in the normal control group was not different from that in SLE patients.CD4+Treg cell count was negatively correlated with SLEDAI score(r=-0.397,p=0.030);positively correlated with C3(r=0.685,p<0.001);positively correlated with C4(r=0.576,p=0.002);and PLT(Platelet)were positively correlated(r=0.435,p=0.016);they were not related to ESR,CRP,WBC,RBC,LY.There was a positive correlation between CD8+Treg cell count and WBC(white blood cell count)(r=0.422,p=0.020),which was not associated with SLEDAI score,ESR,C3,C4,CRP,RBC,PLT,LY.(2)Low-dose IL-2(50WIU/day x 5 days)increased CD4+Treg cells count compared with pretreatment(P<0.001);After treatment,CD8+Treg cells count were higher than those before treatment(P<0.05).Conclusion: 1.There is no significant difference in CD8+Treg cells between patients with systemic lupus erythematosus compared with normal individuals;CD4+Treg cells are reduced compared with normal individuals and are associated with disease activity.2.Low-dose IL-2(50 WIU/day x 5 days)could promote the expression of CD4+ Treg and CD8+ Treg cells in peripheral blood of SLE patients.
