Study On The Pathogenesis Of Qi Deficiency Poison Accumulate Syndrome Of Rheumatoid Arthritis And To Explore The Effect Mechanism Effective Prescription

Objectives: Based on literature research and clinical diagnosis and treatment,the pathogenesis of qi deficiency syndrome of rheumatoid arthritis was preliminary proposed and further explore the mechanism of its effect on the treatment of rheumatoid arthritis,and provide experimental basis and theoretical support for the clinical application of qi deficiency and qi deficiency syndrome of rheumatoid arthritis.Methods:1.Retrieve for rheumatoid arthritis related literature,analysis of its etiology and pathogenesis and clinical manifestations.2.Analysis of clinical cases of rheumatoid arthritis.3.The mechanism of square effect of rheumatoid arthritis: The animal models of adjuvant arthritis were randomly divided into blank group,model group,adjustable square group,yiqi group,detoxification group,yuping wind group:(1)The changes of the general condition,body weight and foot swelling were observed.(2)Enzyme linked immunosorbent assay was used to detect the contents of IL-6 and IL-4 in serum of rats.(3)Pathological changes of synovial tissue were observed under light microscope.(4)The expression level of Foxp3,ROR?t,TGF-? mRNA genes in the differentiation of Th17/Treg cells were detected by PCR.Results:1.Put forward the deficiency of toxic aggregates as the basic pathogenesis of rheumatoid arthritis.2.Proposed the basic pathogenesis of clinical immune imbalance disease for the qi and toxic,adjustable squared as an effective prescription,can be used for the treatment of rheumatoid arthritis.3.The mechanism of square effect of rheumatoid arthritis:(1)Symptom observation: except the blank group,other groups were injected rats after 24 h foot swelling reached the peak,the local temperature was significantly increased,suggesting that the model of AA rats limp was successful.The weight of adjustable square group gained the most,compared with the model group,there are differences(P < 0.05).The treatment group of foot swelling has decreased,the adjustable square foot swelling group,Yiqi group dropped the most,and the other groups except the control group had significant difference(P < 0.05),suggesting that regulation group can reduce the square foot swelling.(2)Immune factor observation: The content of IL-6 in the group was significantly lower than that in the model group(p <0.05).The content of IL-4 in the control group was higher than that in the other treatment groups except the Yiqi group,and the IL-4 content in the Yiqi group was the highest.(3)Pathological observation: synovial tissue in the model group increased infiltration of inflammatory cells,synovial hyperplasia,fibrous tissue hyperplasia and macrophage proliferation,tide line integrity,compared with the blank group,which had significant difference(P < 0.01),suggesting that the model was successful;Compared with the model group,Adjustable square decreased in the proliferation of synovial membrane and the decrease of fibrous tissue proliferation,and there was a difference in the line integrity of the Yiqi group(P < 0.05).(4)Immune related gene expression level: adjustable square group Foxp3 mRNA expression levels were higher than the blank group,which lower than in the model group and Yuping group,detoxification wind close to the blank group,each group had no significant difference in statistics;Compared with the model group,the treatment group ROR?t mRNA expression levels were significantly decreased,detoxification group was the lowest,followed by adjusting square group and Yuping group,there was statistical significant differences(P < 0.05);the expression level of each treatment group TGF-?mRNA increased,there were no significant difference between groups.Conclusion:1.The basic pathogenesis of rheumatoid arthritis is deficiency of qi,which is the effective prescription,which provides a new way for the diagnosis and treatment of rheumatoid arthritis.2.Rheumatoid arthritis effect adjustable square can reduce the local swelling,increase the content of IL-4 and up regulation of anti-inflammatory anti-inflammatory cytokine gene Foxp3 expression level of proinflammatory cytokines;the content of IL-6 and down-regulation of proinflammatory gene ROR?t expression level decreased;inhibiting synovial hyperplasia,fibrous tissue hyperplasia has certain two-way regulation immune trend.
