Objective:To research the effect and mechanism of acupuncture at neiguan regulating Adenosine Receptor on myocardial energy metabolism in myocardial ischemia rats.Methods:240 SPF healthy SD rats were randomly divided into blank control group,sham operation group,model group,acupuncture group,lentivirus infection negative model group,lentivirus infection negative acupuncture group,A1 silencing model group,A1 silencing acupuncture group,A2 a silencing model group,A2 a silent acupuncture group,A2 b silencing model group,A2 b silencing acupuncture group.The myocardial ischemic model and adenosine receptor gene silencing myocardial ischemia model were established by surgically ligating the left anterior descending coronary artery and lentivirus infection to silence the adenosine receptor gene.The acupuncture intervention was performed on the next day after surgery.Secondly,for consecutive 5 days,all acupuncture groups were acupunctured at Neiguan point.The blank control group,sham operation group and each model group were not subjected to acupuncture intervention and were only tied.The rats were sampled and their physiological electrocardiogram,TTC staining,and high performance liquid chromatography were used to detect the electrocardiogram,myocardial infarct size and myocardial ATP,ADP,AMP,and ADO.Result:1.Electrocardiographic changes in each group rats: After myocardial ischemia model,the ST segment of electrocardiogram in rats of each model group was significantly elevated compared with the sham operation group,indicating successful modeling.After treatment,compared with the model group,ST segment of electrocardiogram of rats in acupuncture group was significantly decreased(P<0.001);ST segment of electrocardiogram of lentivirus infection negative acupuncture group was compared with corresponding model group after lentivirus infection.Significantly decreased(P<0.001);A2a silent acupuncture group and A2 b silent acupuncture group was significantly reduced(P<0.001);A1 silent acupuncture group electrocardiogram ST segment was not significantly reduced(P>0.05).2.Myocardial infarct size changes in each group: Myocardial infarct size in each model group was significantly higher than that in the sham-operated group after myocardial ischemia(P<0.001).Compared with the model group after treatment in each group,the myocardial acupuncture group The infarct size was significantly reduced(P<0.001).Compared with the corresponding model group,the infarct size of the acupunctured group with Lentivirus infection was significantly reduced(P<0.001)after acupuncture with lentivirus infection;A2a silent acupuncture group Significantly decreased(P<0.001);A1 silent acupuncture group and A2 b silent acupuncture group did not significantly reduce the myocardial infarct size(P>0.05).3.Changes in Contents of Energy Substances ATP,ADP,AMP and ADO in each group:(1)Changes of myocardial ATP and ADO content: After AMI,the ATP and ADO levels in each model group were significantly lower than those in the sham group(P<0.001).After treatment,compared with the model group,the myocardial ATP and ADO levels in the acupuncture group were significantly increased(P<0.001);after lentivirus infection,the acupuncture group was compared with the corresponding model group in each group.Myocardial ATP and ADO levels were significantly increased in rats(P<0.001);A2a silent acupuncture group was significantly increased(P<0.001);A1 silent acupuncture group and A2 b silent acupuncture group had no significant increase in myocardial ATP and ADO(P>0.05).(2)Changes of Myocardial ADP Content: After AMI,the ADP content in rats of each model group had no significant changes compared with the sham group(P>0.05);after treatment,compared with the model group,the ADP of myocardium in the acupunctured rats There was no significant change in the content(P>0.05).The level of ADP in myocardium was not significantly different between the acupuncture group and the corresponding model group after infection with Lentivirus(P>0.05).(3)Changes in Myocardial AMP Content: After AMI,the AMP content in the rats of each model group was significantly higher than that in the sham group(P<0.001).Compared with the model group after treatment in each group,the acupunctured group rats' myocardium the content of AMP was significantly decreased(P<0.001).Compared with the corresponding model group,the content of AMP in myocardium of rats with lentivirus infection-negative acupuncture was significantly lower after infection with lentivirus(P<0.001);A2a silent acupuncture significantly decreased in the group(P<0.001);A1 silent acupuncture group and A2 b silent acupuncture group myocardial AMP content was not significantly reduced(P>0.05).Conclusion:1.Acupuncture at Neiguan points can produce a good protective effect on myocardial tissue.Acupuncture can significantly inhibit the ST-segment elevation of myocardial electrocardiography in rats,reduce the myocardial infarct size caused by ischemia and hypoxia and optimize myocardial energy metabolism.2.The protective mechanism of myocardial ischemia induced by acupuncture may be through regulating the adenosine A1 receptor and adenosine A2 b receptors to up-regulate the myocardial ATP content,down-regulate the myocardial AMP content and increase the myocardial tissue ADO content,further improves the energy metabolism of myocardial tissue,thereby protecting the myocardial tissue.
