Objective:Based on the Traditional processing literature and traditional processing technology,we found the theoretical basis of the three Rhizoma Chuanxiong processed by honey,Angelica and tea.In the research of processing technology,we optimized it with the orthogonal experiment and response surface method and determined the best processing condition.Moreover,the quality standard of the three kinds of product was studied and a draft of the corresponding quality standard was set up,which lay the foundation of further improvement.At last,the pharmacodynamics of three kinds of Rhizoma Chuanxiong pieces were studied,so as to provide scientific basis for clinical application.Methods:1 the Research of Processing TechnologyThe contents of Ferulic acid andLigustilide in Rhizoma Chuanxiong processed by traditional method were determined by HPLC and used as comprehensive evaluation indexes.the single test and the Box-Behnken design-response surface method(BBD-RSM)were adapted to examine the influence of critical factors on processing technology and we have obtained the best processing condition.2 the Research of Quality StandardAccording to the test method of the Chinese Pharmacopoeia of the 2015 edition,the characters,identification,examination,extract and content of three kinds of Rhizoma Chuanxiong were evaluated.Then,the 11 batch of products were evaluated which processed by the best processing condition.On the basis of experimental results above,we set up the the quality standard and a draft of the corresponding quality standard of the three kinds of product.3 the Research of Pharmacological EfficacyAccording to the different clinical efficacy of each of the three Rhizoma Chuanxiong Pieces,the effects of concocting on the efficacy of the three RhizomaChuanxiong decoction pieces were verified with the acute blood stasis model,the anti-inflammatory analgesia model and the migraine model.Results:1 the Research of Processing TechnologyWe investigated the key factors in the processing of Rhizoma Chuanxiong,such as processing time,processing temperature,the amount of excipient and moistening time.The results showed that the optimum processing technology of Rhizoma Chuanxiong processed with honey consisted of adding 25 g honey to 100 g Rhizoma Chuanxiong,moistening 2.5 h,15 min of processing time at 160 ?.The optimum processing technology of Rhizoma Chuanxiong processed with Radix Angelicae dahuricae consisted of adding 250 g Radix Angelicae dahuricae to 100 g Rhizoma Chuanxiong,8 min of processing time at 140 ?.The optimum processing technology of Rhizoma Chuanxiong processed with Tea consisted of adding 15 g tea to 100 g Rhizoma Chuanxiong,16 min of processing time at 120 ?.2 the Research of Quality StandardWe studied the quality standards of 11 batches of three kinds of Rhizoma Chuanxiong,included the characters,identification,examination,extraction and content determination.Finally,the quality standard draft of three kinds of pieces was determined.3 the Research of Pharmacological EfficacyBoth the Rhizoma Chuanxiong and the Rhizoma Chuanxiong processed with honey have obviously improved the hemorrheology indexes of rats with acute blood stasis syndrome,after processed with honey,relaxed the blood activating effect.The Rhizoma Chuanxiong,the Rhizoma Chuanxiong processed with honey and the Rhizoma Chuanxiong processed with tea all can ease the pain which caused by acetic in mice(P<0.05 or P<0.01).When compared with the the Rhizoma Chuanxiong,the Rhizoma Chuanxiong processed with tea or honey have significant difference in the effect of easing pain in mice.When compared with the the RhizomaChuanxiong,the Rhizoma Chuanxiong processed with tea have significant difference in head incubation period in rat(P < 0.05),increasing the time of head incubation period,but have no significant difference in the number of scratching(P>0.05),but there was a tendency to reduce the number of scratching.The result have indicated that after processed the Rhizoma Chuanxiong,enhanced the effect of relieving pain.Conclusions:We found out the main factors in the processing of Rhizoma Chuanxiong after looking up the literature of the past dynasties.We have determined the specific parameters of the processing of three kinds of Rhizoma Chuanxiong,established the quality standard and the draft of quality standard,to control the decoction pieces.Through the study on the pharmacodynamics of the three kinds of pieces,it was found that the efficacy have improved after the processing.It not only provides a reference for the actual production,but also protects and develops traditional processed pieces,and has certain significance to guide the producing of the processed slices normally.
