ObjectiveTo investigate the roles of JNK MAPK in IL-4-induced M2 macrophage polarization.MethodsRAW264.7 derived M2 macrophage(RAW264.7-M2)was established with 20ng/ml IL-4.The cytokine production of IL-4-stimulated RAW264.7 cells were analyzed by using quantitative PCR and western blot analysis.In addition,quantitative PCR and western blot analysis were also used for determining mRNA and protein levels of JNK and cmyc.Cell migration was examined by scratch wound healing assay while cell proliferation was assessed by 5-Ethynyl-21-deoxyuridine assay.ResultsM0 macrophages can be polarized into M2 status in response to IL-4 stimulation,indicated by the increased expression of the M2-specific cytokine markers Arg1 and Mrc1.JNK MAPK and transcription factors cmyc were up-regulated in IL-4-M2 macrophages.Furthermore,inhibition of JNK suppressed the expression of M2 markers Arg1,Mrc1 and cmyc,Irf4,which also regulated M2 markers.Moreover,we showed that M2 macrophages had increased ability of migration depending on cytokines production.ConclusionsTaken together,our data establishes a new role for JNK MAPK in IL-4-induced alternative activation of macrophages and may provide a novel strategy for immune therapy.
