ObjectiveTo investigate the effect of metformin(MET)on the expression of AQP7 in T2 DM rats and the possible mechanism underlying its protective effect in pancreas.MethodsHigh fat die combined low –dose STZ was induced to T2 DM model 100 male rats were randomly divided into the normal control group and T2 DM model groups.The successful T2 DM model rats were randomly divided into T2 DM model group,T2 DM physiological saline control group,low-dose MET group,middle-dose MET group,high-dose MET group.After MET treatment,serum concentrations of cholesterol(TC),triglycerides(TG)and glucose(GLU)were measured in all studied groups;HE staining was used to identify morphological changes.Immunofluorescence was used to investigate the distribution of AQP7 in rat pancreas tissue.The expression of AQP7 and LEPTIN as well as MAPK was assessed by western blotting.The content of glycerol in pancreas was detected by Glycerol Assay Kits.ResultCompared with the normal control group,the serum concentrations of cholesterol(TC),triglycerides(TG)and glucose(GLU)were increased in T2 DM group(p<0.05),HE results shown that the pancreas cells appeared edema,necrosis and the smaller islet clusters in T2 DM group.The expression of AQP7 was decreased(P<0.05),the ratio of p-JNK/JNK,P-P38/P38 and the expression of LEPTIN were increased,and p-ERK/ERK remained stable in T2 DM group(p<0.05).The content of glycerol in pancreas was increased in T2 DM group detected by Glycerol Assay Kits(P<0.05).MET could reverse the above effects in a dose-dependent manner,especially in middle and high dose group.Conclusion MET treatment could ameliorate pancreatic necrosis and glyeolipid metabolism disorder associated with T2 DM via the up-regulation of expression levels of AQP7.
