Effect Of Herbs Of Supplement Qi And Yin,Eliminating Stasis And Dredging Meridians On Diabetic Gastroparesis Rats And Effect On Variations Of Cyt C,Caspase-9 And Caspase-3 In Rats With Diabetic Gastroparesis

Objective: To research mechanism of diabetic gastroparesis(DGP)and the treatment of herbs which can supplement Qi and Yin,eliminate stasis and dredge meridians.Methods: 50 male Sprague Dawley rats were assigned randomly into control group and model building group.Control group was fed by normal diet,and model building group was established as model which was fed by high sugar and high fat diet before given intraperitoneal injection of streptozotocin.All the models were assigned randomly into model group and therapeutic group.Therapeutic group was given intragastric administration of herbs once a day.Compared with it,control group and model group were given intragastric administration of purified water with the same volum.All the rats would be killed after 16 weeks treatment.Collected gastric content for measuring the rate of gastric emptying.Collected gastric tissue for analyzing histological structure,the rate of apoptosis in gastric smooth muscle cells,the expression of cytochrome c(Cyt c),the activity of cysteinyl aspartate-specific proteinase 9(Caspase-9)and cysteinyl aspartate-specific proteinase 3(Caspase-3).Results: Compared with control group,the rate of gastric emptying in model group and therapeutic group were both significantly lower(P<0.05).Compared with model group,the rate of gastric emptying in therapeutic group was significantly higher(P<0.05).Compared with control group,the rate of apoptosis of gastric smooth muscle cells in model group and therapeutic group were both significantly higher(P<0.05).Compared with model group,the rate of apoptosis of gastric smooth muscle cells in therapeutic group was significantly lower(P<0.05).Compared with control group,the average optical den-sity of Cyt c in model group and therapeutic group were both significantly higher(P<0.05).Compared with model group,the average optical density of Cyt c in therapeutic group was significantly lower(P<0.05).Compared with control group,the enzyme activity of Caspase-9 and Caspase-3 in model group and therapeutic group were both significantly higher(P<0.05).Compared with model group,the enzyme activity of Caspase-9 and Caspase-3 in therapeutic group were both significantly lower(P<0.05).Conclutions: Delayed gastric emptying was found in DGP rats.Herbs of supplement Qi and Yin,eliminating stasis and dredging meridians can improve gastric emptying and contribute to curing DGP.Increased apoptosis of gastric smooth muscle cells was found in DGP rats.The mechanism of DGP may related to mitochondrion mediated excessive apoptosis of gastric smooth muscle cells.The mechanism of the treatment of herbs may related to decreasing mitochondrion mediated excessive apoptosis of gastric smooth muscle cells.
