Objective:Explore the effective extraction and purification methods for the effective parts of Yiqi Huoxue Compound,to observe the inhibition of thrombus formation and its mechanism by the effective fraction of Yiqi Huoxue Compound with the rat model of carotid artery thrombosis.Methods:1.Salvia miltiorrhiza tanshinone IIA and salvianolic acid B were used as indexes,and ethanol was used for reflux extraction.The dregs were evaporated to ethanol and extracted with water again to obtain the fat-soluble and water-soluble components of Salvia miltiorrhiza.The total amount of ginsenosides Rg1,Rb1,and Re in American ginseng was used as an inspection index,and ethanol was used for reflux extraction to obtain the fat-soluble components of American ginseng.The macroporous resin separation method was used to separate the effective parts and the effective parts of Salvia miltiorrhiza and American ginseng were obtained.The effective parts of Salvia miltiorrhiza and American ginseng were uniformly mixed in a ratio of 3:1 to produce Yiqi Huoxue compound.2.Fifty SD male rats were randomly divided into sham operation group?S?,model group?M?,aspirin group?A?,Yiqi Huoxue compound high?H?and low?L?dose groups.After 7days of prophylaxis,sham operation group was given normal saline and the other groups were given thrombosis induced by FeCl3 on topical carotid artery.The rats were intragastrically administrated for 21 days and were detected by enzyme-linked immunosorbent assay?E-Lisa?.The contents of TXB2,6-K-PGF1?,AT-?,PC,PLG,,Endothelin-1?ET-1?were measured.The blood vessels of carotid artery thrombosis part were taken after the blood was collected,and the pathological sections were analyzed by HE staining.Results:1.Apply water extraction,ethanol reflux extraction and macroporous resin separation and other methods to optimize the process of extraction and purification.Then make into Yiqi Huoxue Compound with Salvia and American ginseng.2.By optimizing the extraction and purification methods of Salvia miltiorrhiza and American ginseng,the preparation cycle of replenishing qi and promoting blood circulation is shortened,and the efficiency of compound preparation is improved.3.A rat model of carotid thrombosis was established by external carotid artery-induced thrombosis with FeCl3,and visual thrombosis was observed in the M group.4.Compared with S group,The platelet aggregation rate,TXB2 content,TXB2/6-K-PGF1?ratio,and ET-1 content were increased?P<0.05?;the contents of 6-K-PGF1?,PC,AT-III,and PLG were decreased in group M?P<0.05?.The difference was statistically significant.5.Compared with group M,the contents of 6-K-PGF1?,PC,AT-III and PLG were increased?P<0.05?;TXB2 content and TXB2/6-K-PGF1?ratio were significantly decreased in H and L groups?P<0.01?;The ratio of TXB2,PLG and TXB2/6-K-PGF1?were significantly decreased?P<0.01?,and the content of 6-K-PGF1?was significantly increased in group A?P<0.01?.The difference was statistically significant.6.Compared with group M,although the content of ET-1 in group H and L decreased,it did not show statistical difference.7.Pathological sections showed that there was thrombosis in the common carotid artery of rats after external application of FeCl3,and aspirin and Yiqi Huoxue compound all reduced the thrombus to varying degrees.Conclusion:1.The method of thrombosis induced by Fe Cl3 on topical carotid artery has a high success rate and the result is easy to observe.It can be used as a examiningmodel for the experiment of Yiqi Huoxue compound.2.Salvia miltiorrhiza and American ginseng are combined in a ratio of 3:1 for Yiqi Huoxue compound,which has an inhibitory effect on carotid artery thrombosis and may promote the elimination of thrombus.Its mechanism may be through the reduction of plasma TXB2,and the growth of 6-K-PGF1?.Its mechanism may be through the growth of PC,AT-?,thus enhancing the activity of anticoagulant system.It is associated with increased PLG content and activity of the fibrinolytic system.
