Objective To explore the roles of NF-?B pathway in the changes of cytokines and cytotoxicity treated with the rare earth neodymium oxide particles in rat alveolar macrophages(NR8383)cell line.Methods NR8383 cells in the logarithmic growth phase were exposured to final concentrations of 0(control,PBS),1.56,3.125,6.25,12.5,25,50,100,200 ?g/mL of neodymium oxide suspension for 12,24,48 h,MTT methods was used to assay the cell inhibition rate.NR8383 cells in the logarithmic growth phase were exposured to final concentrations of 0(control,PBS),1.56,3.125,6.25,12.5,25,50,100?g/mL of neodymium oxide suspension for 24 h,the content of TNF-?,IL-1?,MIP-1?,MCP-1,TGF-?1 and IL-8 on NR8383 cells were measured by ELISA methods.NR8383 cells in the logarithmic growth phase were exposured to final concentrations of0(control,PBS),1.56,3.125,6.25,12.5,25,50?g/mL of neodymium oxide suspension for 24 h,p-NF-?Bp65,p-IKK? and Caspase-3 protein on NR8383 cells were measured by Western blotting method.Results Compared with the control group,NR8383 cell inhibition rate were higher to exposure the concentration of neodymium oxide at different times after,Wherein the concentration of 1.56,3.125,6.25?g/mL inhibition rate of the three groups showed no significant difference(P >0.05),Other inhibition rate at each concentration difference was statistically significant(P <0.05).And concentration of TNF-?,IL-1?,MIP-1?,MCP-1,TGF-?1 and IL-8 in NR8383 cell supernatant were higher to exposure each concentration of neodymium oxide,had significant difference(P<0.05).NR8383 cell inhibition rate and the concentration of TNF-?,IL-1?,MIP-1?,MCP-1,TGF-?1 and IL-8 in NR8383 cell supernatant significantly increased with the increase of the concentration of neodymium oxide.NR8383 inhibition rates were positively correlated with the concentration of TNF-?,IL-1?,MIP-1?,MCP-1,TGF-?1and IL-8 in NR8383 cell supernatant(P <0.01).Concentrations of TNF-?,IL-1?,MIP-1?,MCP-1,TGF-?1 and IL-8 in NR8383 cell supernatant were positively correlated between the two(P <0.01).The expressions of Caspase-3,p-NF-?Bp65,p-IKK? protein in NR8383 cell were positively correlated with the concentration of TNF-?,IL-1?,MIP-1?,MCP-1,TGF-?1 and IL-8 in NR8383 cell supernatant(P<0.01).Expressions of Caspase-3,p-NF-?Bp65,p-IKK? protein in NR8383 cell were positively correlated between the two(P<0.01).Conclusions Rare earth neodymium oxide particles caused rat alveolar macrophages(NR8383)inflammatory injury,causing chemokines cytokines and inflammatory cytokines in NR8383 cell supernatants.And the expression of p-IKK?and p-NF-?Bp65 in NR8383 cell was significantly increased,showed that NF-?B signaling pathway is activated.It suggests that NF-?B signaling pathway may be one mechanism of inflammatory injury treated with the rare earth neodymium oxide particles in rat alveolar macrophages(NR8383)cell line.
