| Objective: To investigate the protective effect of PON1 on brain tissue of DDVPpoisoned rats.Method: Thirty male Sprague Dawley rats were randomly divided into 5 groups(6 rats / group),A group(control group),B group(exposure group),C group(PON1 group),D group(Traditional-therapy group),E group(combination-therapy group).Group A: Rats were injected intraperitoneally with 1 ml/kg physiological saline.Group B : Rats were intraperitoneal injected by DDVP(15mg/kg).Group C: Rats were injected with rabbit serum PON1 9600U/ kg by caudal vein 30 min before intraperitoneal injection of 15mg/kg DDVP(shaken thoroughly).D group: rats intraperitoneal injected 15mg/kg DDVP(shaken thoroughly)immediately given atropine 10mg/kg+pralidoxime iodide(45m/kg).Group E: Rats were intravenously injected with rabbit’s serum PON1 9600U/kg 30 min before injection of 15mg/kg DDVP(shaken thoroughly)and immediately intraperitoneal injected of atropine 10mg/kg+ pralidoxime iodide(45m/kg).The rats in each group were sacrificed 8 h after model establishment.Results: Serum choline esterase(CAT)and cortisol(CAT)activity were detected by microplate reader.The expression of Sod1,Pink1 and Prdx6 in cerebral cortex and brain was detected through western blot.HE staining was used to observe the pathological changes in the cerebral cortex and hippocampus.The serum levels of Ch E and CAT in group B decreased(p <0.05)compared to group A,and the serum Ch E and CAT in group D was decreased(p <0.05)compared to group B while was increased compared to group C(p < 0.05).The serum level of Ch E and CAT in group E was decreased compared to group A,but there was no statistical significance(p >0.05).The expression of Sod1,Pink1 and Prdx6 in the cerebral cortex and hippocampus from group B were the lowest(P<0.05)compared with the other 4 groups.The expression of Sod1,Pink1,Prdx6 in group C was lower compared to group A,while higher than that in group B(P<0.05).The expression of Sod1,Pink1 and Prdx6 in group D was the lowest compared to group A and group C,while higher than group B(P<0.05).The expression of Sod1,Pink1 and Prdx6 in group D is less than that in group A,but higher than the other 3 groups.The expression of Sod1,Pink1 and Prdx6 among group A,B,C,D and E is almost the same without statistical significance(P>0.05).Serious cell edema was observed under light microscope.Nuclear condensation,vacuolization and steat osis were obvious.The damage in group C was alleviated than that in group B,and the damage in group D was reduced than that in group C.The damage in group E was the slightest among the group B,C and D,nevertheless,which was more severe compared with group A.There was no significant difference in damage between cerebral cortex and hippocampus.Conclusion: Rabbit serum had a protective effect on oxidase 1 against cerebral cortex and hippocampus in rats exposed to dichlorvos. |
