Objective: The pathogenesis of developmental dysplasia of the hip(DDH)are numerous and difficult to clear at present.Previous researches and observations confirm that the change of acetabulum is closely related to it.This research compared the differential expression of asporin(ASPN)on the m RNA and protein level in the articular cartilage between the normal and DDH children,in order to explore what cause the change.of acetabulum cartilage in DDH.Materials and methods: We collected some acetabulum cartilage tissue as the research object,which came from those DDH patients who had a hip open reduction treatment in shengjing hospital affiliated to China medical university from 2013 to 2016(a total of 26 cases,30 hips,23 cases were female and 3 cases were male,ages ranged from 6 months to 6 years old,average 2.42±1.51 years old).We also collected some acetabulum cartilage tissue as the normal controls,which came from the children who with fetal amputation for malignant tumor induced labor or death from other nonarticular diseases and so on(a total of 10 cases,15 hips,6 cases were female and 4 cases were male,ages ranged from 0 months to 5 years old,average 0.33±1.25 years old).All above organizations are saved with liquid nitrogen or in-80 ? condition,and to detected the expression of ASPN at protein and m RNA level with those methods,including in i TRAQ(isobaric tags for relative and absolute quantitation),Western-blot and RT-PCR.Using SPSS software(version 21.0,the United States)for statistical data analysis,data are used to mean + standard deviation(x + s)way,using two sample t test method to carry on the comparative analysis between groups,using multiple linear regression method to evaluate the correlation between variables,P < 0.05 as a statistically significant difference.Result: 1.The result of iTRAQ experiment showed that: the expression of ASPN were increased in the DDH group compared to normal group(1.10 ± 0.04(DDH group)vs.0.93± 0.03(normal group),the differences were statistically significant(p<0.05).2.The result of Rt-PCR experiment showed that: the expression of ASPN at m RNA level were increased in the DDH group compared to normal group(2.69±0.52(DDH group)vs.1.00±0.44(normal group),the differences were statistically significant(p<0.05).3.The result of Western-blot experiment showed that: the expression of ASPN were increased in the DDH group compared to normal group(1.53±0.36(DDH group)vs.0.34±0.07(normal group),the differences were statistically significant(p<0.05).4.The result of correlation analysis showed that:no correlations were found between ASPN gene expression and age or gender(p=0.44,p=0.06).No correlations were found between ASPN protein expression and age or gender(p=0.58,p=0.23).Conclusion: The expression of ASPN appeared significantly raised that compared to the normal in the acetabular cartilage tissue of DDH,and those change may be related to the abnormal form of the acetabulum in DDH.In other words,the differential expression of asprin may be one of the pathogenic factor of DDH.
