Objective: Diabetic kidney disease(DKD)is one of the more serious complications in people with type 2 diabetes mellitus(T2DM).Without early intervention,it may cause renal damage or even renal failure.It is a key cause of end-stage renal disease.Early diagnosis and prevention of DKD reveals that specific and sensitive laboratory serological markers are crucial for patients with diabetes.We aim to study the expression of forkhead transcription factor O1(FOXO1)in patients with type 2diabetes mellitus in different urinary albumin excretion stage and to explore the correlation with osteopontin(OPN)and other serum biological indicators.Methods: The subjects were type 2 diabetes patients and they all met the conditions for newly diagnosed in our hospital.According to the urinary albumin / creatinine ratio(UACR): normal albuminuria(UACR <30mg / g,n = 126),microalbuminuria(UACR: 30-300 mg / g,n =118),microalbuminuria hypertensive(UACR: 30-300 mg/ g with hypertension,n =102),clinical albuminuria(UACR> 300 mg / g,n =72)clinical albuminuria hypertensive(UACR> 300 mg / g and with hypertension,n =88).Control group selected the age,gender-matched healthy people.Fork head transcription factor O1(FOXO1)and osteopontin(OPN)were measured by ELISA.Results The levels of FOXO1 and OPN in normal albuminuria,microalbuminuria,microalbuminuria hypertensive,clinical albuminuria and clinical albuminuria hypertensive were significantly higher than those in control group(P <0.05).Results: Serum FOXO1 levels were positively correlated with body mass index,UACR,TG,uric acid,Hb A1 C,HOMA-IR,systolic blood pressure,diastolic blood pressure and OPN levels(r = 0.378,0.521,0.385,0.396,0.444,0.414,0.629,0.516,0.342,all P <0.01),and had a significant negative correlation with HDL-C(r =-0.345,P<0.01).Conclusions: FOXO1 is expected to become a new serum marker for early detection of DKD and evaluation of renal lesions.Serum FOXO1 might be a new direction for the treatment of diabetic nephropathy.
