Objective: To investigate the role of Ras family member A(Rho A)in ovarian cancer cell SKOV3/DDP drug resistance.Methods: set up the experimental group and control group,the expression of Rho A silenced SKOV3/DDP cells by si RNA interference in the experimental group,the control group set up negative control(si-Scramble RNA),mechanism of changes of cell cycle and apoptosis detection cell phenotype and may exist,the expression of Western Blot detection of Rho A,and the expression of the experimental group and the drug resistance of ovarian the control group in cancer cells and apoptosis related protein,MTT to detect different concentrations of cisplatin on the experimental group and control group of ovarian cancer cell inhibition rate changes,changes in drug sensitivity to cisplatin by SKOV3/DDP cells was observed after Rho A interference and related protein levels of drug resistance related protein,apoptosis,to investigate the role of Rho A in the process of drug resistance of ovarian cancer the probablemechanism.Results: the results of Western blot showed that the expression level of Rho A in SKOV3/DDP cells was significantly higher than that of SKOV3 cells.MTT detection and suppression of cisplatin with different concentrations of the two groups of tumor cell rate increased with the dose of cisplatin,the survival rate of tumor cells in the two groups decreased gradually,at 300 ng/ml,600 ng/ml,1200 ng/ml,2400 ng/ml,4800 ng/ml concentration,the survival rate of the experimental group was significantly lower than the control group(P < 0.05).The results suggest that SKOV3 /DDP interference Rho A,increased sensitivity to cisplatin in ovarian cancer cells,cell cycle test results show that,compared with the control group,after SKOV3/DDP cells transfected with si RNA Rho A G2/M phase arrest and apoptosis.The results showedthat compared with the control group,the apoptosis increased after SKOV3/DDP cells were transfected with si RNA Rho A,inhibited ABCG1,Bcl-2 and surviving protein expression.Conclusion: Rho A involves in ovarian cancer cell durg resistance through modulating ABCG1,Bcl-2 and surviving...
