Objective:Sepsis,a systemic inflammatory response syndrome triggered by various infectious agents invade the body,involves both innate and adaptive immunity.Macrophages,a kind of phagocytes distributed in almost all tissues of the body,play an important role not only in innate immunity,but also innate and adaptive immunity.Macrophages'differentiation and its function is particularly important in the sepsis.Hyperbaric oxygen therapy as an effective treatment for sepsis has been confirmed.After hyperbaric oxygen therapy,the differentiation and function of macrophage must change,but it is unclear.The present study explored the changes of peritoneal macrophage function in sepsis mice after hyperbaric oxygen therapy.Methods:1?One hundred C57BL/6 mice were randomly averagely allocated into the following four groups:A(normal control group)(normal saline+air),B(sepsis group)(zymosan+air),C(hyperbaric Oxygen group)(normal saline+hyperbaric oxygen),D(sepsis+hyperbaric oxygen group)(zymosan+hyperbaric oxygen);the sepstic mice were treated by intraperitoneal injection with zymosan,groups B and D were given intraperitoneal injection of zymosan and groups A and C were injected with equal volume of saline,mice in groups C and D were respectively fed with 100%pure oxygen(2ATA)at 4 hours and 11 hours after the start of the experiment for 2 hours.2?The number of surviving mice was obsevred at 72h after the intraperitoneal injection with zymosan;At the 18th hour after the start of the experiment,the peripheral blood of the mice was taken and the changes of the inflammatory cytokines in the peripheral blood of mice were detected by enzyme-linked immunosorbent assay(ELISA);At the 18th hour after the start of the experiment,macrophages were isolated by immunomagnetic bead and the number of M1 macrophages was determined by flow cytometry;The expression of mRNA of inducible nitric oxide synthase(iNOS)in Peritoneal macrophage was detected with RT-PCR;3?Statistical analysis methods:SPSS 19.0 statistical software for statistical analysis,the measurement data were expressed as mean±standard deviation((3?±),one-way ANOVA(LSD test for variance and Dunnett's T3 test for variance)were used for the comparison among groups,?~2Fisher test was used for the survival of the mice,P<0.05 for the difference was statistically significant.Results:Compared with sepsis group,The number of surviving mice of sepsis+hyperbaric oxygen group was significantly higher(P<0.05);The percentages of M1macrophage in sepsis+hyperbaric oxygen group was significantly lower than that in sepsis group(P<0.05);the expression of mRNA of inducible nitric oxide synthase(iNOS)in sepsis+hyperbaric oxygen group were also significantly lower than that in sepsis group(P<0.05);The level of IL-12 in peripheral blood in sepsis+hyperbaric oxygen group was significantly lower than that in sepsis group(P<0.05),the level of IL-10 was significantly higher than that in sepsis group(P<0.05).Conclusions:Hyperbaric oxygen therapy had effective effect for increasing the number of surviving mice of sepsis group,reducing the level of IL-12 in peripheral blood and rising the level of IL-10 in peripheral blood;By inhibiting the expression of M1macrophage and iNOS,it can reduce the excessive inflammatory response and play a role in the protection of mice.
