Objective:Atherosclerosis(AS)is an inflammatory disease in which aggregations of lipids and leukocytes become deposited on the inner arterial walls to an extent that threatens human health.A recent study of chronic diseases in China revealed that 33%of adults in China have carotid atherosclerotic plaques.With age,carotid intima-media thickness in Chinese populations is significantly faster than in the UK.If it were possible to estimate the likelihood that an individual has carotid atherosclerosis,it may also be possible to advise a susceptible population on ways to change their life style and diet so as to prevent carotid atherosclerosis and reduce the probability of suffering a stroke.Scavenger receptor Class B CD36 and Class A MARCO play an important role in atherosclerosis.Whether the single nucleotide polymorphisms of these two receptor genes have an impact on susceptibility to carotid atherosclerosis or not,we used a case-control study to investigate the problemMethods:1.Study population:We chose 215 carotid atherosclerosis patients from China Medical University from March 2016 to November 2016 as case group,and we chose 252 cases from the Medical Examination Center of The First Affiliated Hospital of China Medical University as control group in the same period.2.Genomic DNA extraction:Qiagen nucleic acid extraction kit was used to extract genomic DNA from whole blood samples.CD36 and MARCO gene SNP detection:HRM-PCR method was used to detect the subjects MARCO,rs12998782 and CD36 rs1761667,and the results were analyzed using LightCycler 480 Gene Scanning software.4.statistical analysis:We performed statistics using SPSS17.0.The genotype distribution of the control group was checked by Hardy-Weinberg equilibrium.The difference of genotype distribution between the case group and the control group was X~2 test.A two-tailed P value less than 0.05 was considered statistically significant.Results:1.Association of the rs1761667 and rs12998782 polymorphism with carotid atherosclerosis susceptibility:There was no significant difference in genotype distribution between the case group and control group(p>0.05),or between the male case group and control group(p>0.05).Among females,rs1761667 GA was slightly more prevalent in the case group than the control group,while the frequency of GA+AA was slightly higher in the case groups(GA+AA vs.GG)(p<0.05).The frequency of rs12998782 CT was significantly higher in the case group than in the patient group,while the T and CT+TT alleles were significantly more prevalent in the case groups(CT+TT vs.CC)(p<0.01).2.The combined effect of CD36 rs1761667 and MARCO rs12998782 on carotid atherosclerosis in females:We next combined rs1761667(from CD36)with 12998782(from MARCO),to obtain nine combination genotypes,respectively(Table 4).The case groups with rs1761667 GA,rs12998782CT,and with rs1761667 AA,rs12998782 CT had significantly higher gene frequencies than the rs1761667 GG,rs12998782 CC group(P<0.05).Females who carried rs1761667 GA,rs12998782 CT and rs1761667 AA,rs12998782 CT were3.455-fold and 13.412-fold more likely to have carotid atherosclerosis,respectively,when compared with females who carriedrs1761667GG,rs12998782CC.3.Relationship between menopausal status and the rs1761667 and rs12998782polymorphisms:In the postmenopausal female group,the frequency of rs1761667 GA was significant higher in the case group than in the control group,while GA+AA was significantly more prevalent in the case group(GA+AA vs.GG)(p<0.01).The A allele was found more often in patients than in the control group(p<0.05).Rs12998782 CT was more prevalent in the case group than in controls,while the T allele and CT+TT were significantly more prevalent in the case groups(CT+TT vs.CC)(p<0.01).Conclusions:Female patients of genotype GA for CD36 rs1761667 and CT for MARCO rs12998782 were at an increased risk for carotid atherosclerosis.The presence of rs1761667 GA and rs12998782 CT may increase the risk for carotid atherosclerosis among postmenopausal females.
