Expression Of Inhibitor Of Apoptosis Protein Livin In Renal Interstitial Fibrosis

Objective:To observe expression of inhibitor of apoptosis proteins Livin in renal interstitial fibrosis Methods:(1)HK-2 cells were treated with various dose of TGF-β1(0,l,2,5 and 10ng/ml)for 48 h.Phase contrast microscopy was performed to investigate the morphological changes in cells treated with TGF-β1.The expression of Livin protein in each group at different doses was observed by Western blot experiment.(2)24 CD1 mice were randomly divided into Contral group(groupⅰ)and UUO model group(groupⅱ),twelve mice in each group.The day before operation,groupⅰand groupⅱhad been given equal saline lavage.After forteen days,the obstructive kidney was obtained from the postoperative mices and made into slices.Then the expression of E-Cad、Livin and α-Sma were deteced with Immunohistochemical method.Results:(1)Phase contrast microscopy showed fibroblast-like morphological changes in HK-2 cells treated with TGF-β1.Cells disassociated with surrounding cells and were larger and more elongated than control cells.There was little content of Livin in HK-2 cells,but Livin was expressed most in HK-2 cells induced by TGF-β1 at a dose of 2(ng/ml)and at the time of 48 hours,the differences were significant statistically significant(P <0.001).(2)Compared with groupⅰ,the expression of E-Cad in renal tissue reduced,but α-Sma and Livin increased,the differences were statistically significant(P <0.05).Conclusion:Livin is associated with renal interstitial fibrosis,but the causation needs to be explored more.
