Objective: To use human circ RNA microarray to screen the differential expression of circ RNA of plasma between H-type hypertensive patients and normal controls and to study of circ RNA expression profile.The obvious expression differences of circ RNA was verified by RT-PCR.To use bioinformatics method to analy the target genes the signal pathway and the potential regulatory mechanism of the key circ RNA.This will lay the foundation for the futher explore circ RNA as a new diagnosis biomakers of H-type hypertens and the potential therapentic targets.Methods: We select 3 cases of H type hypertension patients and 3 cases of normal healthy controls in the qualified plasma samples as experiment object.The total RNA of plasma between H-type hypertension patients and healthy individuals was obtained by Trizol reagent.Use circ RNA chip to filtrate the differential expression of circ RNA in serum of each experimental group.Afterwards,the otherness results was verified by RT-PCR technology with 10 cases of H type hypertension patients and 10 cases of healthy persons The target genes and the signal pathways associated with circ RNA of mi RNAS were analyed by bioinformatics.The further enrichment of m RNA date were obtained by gene ontology analysis and KEGG pathway analysis.Results: The differentially expression circRNA spectrum of plasma of patients with H-type hypertension were acquired by circ RNA microarray.Compared with healthy controls,the H type hypertension has significant differences in expression of 117 circRNAs,including 74 up-regulated and 43 down-regulated.Three up-regulated(hsa-circ-0091692?hsa-circ-0087631? hsa-circ-0027719)and two down-regulated(hsa-circ-0001065?hsa-circ-406828)were verified by RT-PCR technology.The results are in conformity with chip screening consequence.The outcome revealed hsa-circ-0087631 was up-regulated consistent with the chip screening results.However,the expresslevels of hsa-circ-0091692? hsa-circ-0027719?hsa-circ-0001065 and hsa-circ-406828 were no significant differences between H type hypertensive group and normal control group.The results showed that three 36 mi RNAs and 2198 corresponding target genes by bioinformatics.The signaling pathways involved in circRNAs regulated target genes and the targets of circRNAs may involve in biological process through GO analysis and KEEG analysis.Conclusion:(1)Circ RNAs are abundant in H-type hypertensive patients and normal controls in plasma,We obtained the circRNAs differential expression profile in H-type hypertension and verified that the differential expression of hsa-circ-0087631 in H-type hypertension group is up-regulated.The hsa-circ-0087631 may be closely related to the development of H-type hypertension.(2)GO and KEGG pathway analysis showed that differentially expressed circRNAs may be play an important role in pathogenesis and development of H-type hypertension.(3)The circRNAs-micro RNA network diagram suggests that hsa-circ-0087631 may be provides a new direction for the research of the pathogenesis of H-type hypertension.
