Objective: To observe the effect of Cordyceps sinensis' s extract on the transdifferentiation of renal tubular epithelial cells induced by cyclosporine A(Cs A),and to explore the effect of Cordyceps sinensis' s extract on the m TOR signaling molecular proteins associated with renal tubule EMT.Methods: NRK-52 E was cultured in vitro and divided into 8 groups:Group A:normal control group;Group B: Cs A group;Group C:Cordyceps sinensis' s extract group;Group D:Rapamycin group;Group E: Cs A+Cordyceps sinensis' s extract,Group F: Cs A+Rapamycin;Group G:Cordyceps sinensis' s extract+Rapamycin;Group H: Cs A+Cordsinen sisyceps' s extract + Rapamycin.The viability of NRK-52 E was detected by CCK8,and The expression of ?-smooth muscle actin(?-SMA),Fibroblast specific protein-1(FSP-1)protein and m RNA were detected by RT-PCR and immunohistochemistry,phosphorylated mammalian rapamycin target protein(p-m TOR)were detected by RT-PCR.Results: 1.There was no significant difference in cell survival rate between group A and group C(P>0.05),and all other groups were down-regulated(P<0.05);The cell survival rate in group B and F was significantly lower than that in group D,E,G(P<0.05).There were no significant difference in group D,E,G(P>0.05);Group F was significantly lower than group B(P<0.05);group H was significantly lower than group G(P<0.05).2.Expression of ?-SMA and FSP-1 protein:?-SMA:There was no significant difference in the expression of ?-SMA protein in group A,C,D,G(P>0.05);The expression of ?-SMA protein in group B,E,F,H were significantly up-regulated(P<0.05);The expression of ?-SMA protein were significantly up-regulated in group B and F than that in group E and H(P<0.05);However,there was no significant difference between the group B and group F(P>0.05).FSP-1:There was no significant difference in the expression of FSP-1 protein in group A,C,D,G(P>0.05);The expression of FSP-1 protein in group B,E,F,H were significantly up-regulated(P<0.05);The expression of FSP-1 protein was significantly up-regulated in group B and F than that in group E and H(P<0.05);However,there was no significant difference in ?-SMA protein between the group B and group F(P>0.05);The expression of FSP-1 protein in group B was lower than that in group F(P<0.05).3.?-SMA,FSP-1 and p-m TOR m RNA:?-SMA,FSP-1:There was no significant difference in the expression of ?-SMA,FSP-1m RNA in group A,C,D,G(P>0.05);The expression of ?-SMA,FSP-1 m RNA in group B,E,F,H were significantly up-regulated(P<0.05);The expression of ?-SMA and FSP-1 m RNA was significantly up-regulated in group B and F than that in group E and H(P<0.05);The expression of ?-SMA and FSP-1 m RNA in group B was lower than that in group F(P<0.05).p-m TOR m RNA: The expression of p-m TOR m RNA in group B,E,F,H were significantly up-regulated(P<0.05);The expression of p-m TOR m RNA was significantly up-regulated in group B and F than that in group E and H(P<0.05),group B was lower than group F,and group E was lower than group H.Conclusion: Cordyceps sinensis' s extract may inhibit Cs A induced renal tubular EMT by inhibiting the activity of m TOR signaling pathway.
