Preliminary Study On The San Cai Lian Mei Granules Effect Of Early Renal Function On Type 2 Diabetic Nephropathy In Mice

Objective: In this study,the model of type 2 diabetic nephropathy was induced in C57BL/6J mice by using Streptozotocin(STZ)and feeding with high fat diet,so that we can investigate whether the compound preparation of traditional Chinese medicine San Cai Lian Mei granules can protect the renal function and improve renal injury of mice in the development of diabetic nephropathy,and provide a solid experimental basis for clinical treatment.Methods: The model of type 2 diabetic mice(C57BL/6J mice)was established by intraperitoneal injection of small dose streptozotocin(STZ)plus high fat feed.The successful model mice were randomly divided into the normal group,DN model group,San Cai Lian Mei granules in high,medium and low dose group.After 4 weeks of experiment,fasting blood glucose,body weight,relative kidney weight,UM-ALB(urine microalbumin),UCr(urine creatinine),UUACR(urinary albumin creatinine ratio)and renal pathological changes were detected in mice.Results: 1.Fasting blood glucose: Compared with the normal group,the blood glucose in the DN model group was significantly higher(P < 0.05).Compared with the DN model group,the San Cai Lian Mei granules in high?middle?low dose group mice significantly reduced fasting blood glucose(P < 0.05).2.Weight and relative renal weight:Compared with the normal group,the weight and relative renal weight of the mice in the DN model group increased significantly(P < 0.05).Compared with the DN model group,San Cai Lian Mei granules in high and middle dose group weight and relative kidney weight decreased,the difference was statistically significant(P < 0.05);the body weight and relative kidney weight of the low dose group were also reduced to some extent,but the difference was not statistically significant(P > 0.05).3.The levels of M-ALB,UCr and UACR in mice : Compared with the normal group,the urine M-ALB,UCr and UACR of the mice in the DN model group were significantly increased(P < 0.05).Compared with the DN model group mice,San Cai Lian Mei granules three dose group mice urinary M-ALB level were decreased(P < 0.05);The level of UCr in the high and middle dose groups was reduced(P < 0.05),but there was no significant difference between the low dose group and the modle group(P > 0.05);In the high-dose group,the level of UACR in urine decreased significantly(P < 0.05),and there was no significant difference between the UACR level in the middle and low dose groups compared with the DN model group(P > 0.05).4.Renal pathology: In normal group mice,there is no abnormal structure of glomeruli and renal tubules;no obvious inflammatory cell infiltration,glomerular hypertrophy and mesangial hyperplasia.In the DN model group,the glomerular mesangial cells were proliferated and some of the renal tubules were necrotic and shedding,and a small amount of inflammatory cells were infiltrated in the interstitium.San Cai Lian Mei granules in high dose group showed a small amount of fibrous tissue hyperplasia,and some of the epithelial cells were necrotic and shedding,and the glomerular structure was basically normal.In middle and low dose group,glomerular mesangial cells proliferated,a part of renal tubular epithelial cells were degenerated and necrotic and shedding,but there was no obvious infiltration of inflammatory cells in interstitium.Conclusions: San Cai Lian Mei particles can not only reduce type 2 diabetic nephropathy mice fasting blood glucose,body weight and relative kidney weight,but also reduce the urine M-ALB,UCr and UACR levels,improve the renal pathological changes.It has a protective effect on renal injury in mice with type 2 diabetic nephropathy caused by STZ and high fat feed.
