Therapeutic Effects Of Fusu Mixture On Sepsis Associated Acute Respiratory Distress Syndrome (ARDS) Rats Induced By Endotoxin

Research objective: To evaluate the therapeutic effects of Fusu mixture on sepsis associated acute respiratory distress syndrome(ARDS)rats,by comparing its effects on the levels of TNF-?,HPA,and IL-10 in the serum,and the lung morphological changes of the sepsis associated ARDS model rats.Research methods: A total of 72 male Wistar rats were randomly assigned into five groups,including high dose Fusu mixture group,middle dose Fusu mixture group,low dose Fusu mixture group,model group,and blank group.Rats in the former four groups were injected by 3mg/kg endotoxin through the left femoral vein in 5 minutes to induce sepsis associated ARDS,whereas rats in the blank group were injected by equal volumes of saline in the same way.Two hours later,equal volume of Fusu mixture in different concentrations was given two times by gavage to rats in the high,middle,and low dose of Fusu mixture groups,with dosing interval of 24 hours,and equal volume of saline was given to rats in the model and blank groups in the same way.At 24 and 48 hours after administration,we observed the general conditions of rats in each group,and took randomly half of the rates from each group to observe the pathological changes of lung tissues by dyeing HE method and histochemistry immunity method,and check the levels of TNF-?,HPA,and IL-10 in the rat serum by enzyme linked immunosorbent assay(ELISA).Then we evaluated the therapeutic effects of Fusu mixture on sepsis associated ARDS rats.Research results:(1)Rats general conditions: the general conditions of the rats in the Fusu mixture groups are better than those of the model group.(2)Pulmonary microvessel counts in the visual field observed by histochemistry immunity method: the mean microvessel counts of rats in the blank group ranged between 7 and 9 in the visual field.At 24 hours after administration,the mean microvessel counts of rats in the Fusu mixture groups and model group were lower than those of the blank group,while those in the Fusu mixture groups were greater than the model group.At 48 hours after administration,the blood vessels of rats in the model group were severely destroyed,and the mean microvessel counts of rats in the Fusu mixture groups increased.(3)Lung injury scores evaluated by dyeing HE method: At 24 hours after administration,the lung injury scores of the model group rats increased from 0.91 to1.58.The lung injury scores of the high,middle,and low Fusu mixture groups decreased to 0.916,0.916,and 1.33,respectively.At 48 hours after administration,the lung injury scores of the model group rats increased from 0.91 to 1.83.The lung injury scores of the high,middle,and low Fusu mixture groups decreased to 0.83,1.08,and 1.25,respectively.(4)At 24 hours after administration,the HPA levels in the rat serum of the model group did not show obviously increase when compared with those of the blank group(P>0.05).The HPA levels in the rat serum of the Fusu mixture groups did not show obviously decrease than those of the model group(P<0.01).At 48 hours after administration,the HPA levels in the rat serum of the model group did not show obviously decrease when compared with those of the blank group(P>0.05).The HPA levels in the rat serum of the Fusu mixture groups showed obviously decrease than those of the model group(P<0.01).(5)At 24 hours after administration,the TNF-? and IL-10 levels of the rat serum of model group displayed significantly increase than the blank group(P<0.01).At 48 hours,the TNF-? and IL-10 levels of the rat serum of the Fusu mixture groups exhibited significantly decrease when compared with the model group(P<0.01).No clear difference has been observed in the TNF-? and IL-10 levels of the rat serum among the Fusu mixture groups(P>0.05).Conclusion:(1)Fusu mixture can improve the general condition and life quality of the sepsis associated ARDS model rats.(2)Fusu mixture can relieve the bleeding,inflammation,exudation,and microvessel injury,and improve lung morphology;(3)At 24 and 48 hours after administration,the TNF-? and IL-10 levels of the rat serum of the Fusu mixture groups were significantly decreased.At 48 hours after administration,the HPA levels of the rat serum of the middle and low Fusu mixture groups were obviously decreased.In conclusion,the Fusu mixture has definite therapeutic effect on sepsis associated ARDS.
