Objective:In this study,we will investigate the process of early brain injury after SAH which induced by apoptosis.of endoplasmic reticulum stress.We also demonstrate that Neuritin alleviates early brain injury by inhibiting the apoptosis which induced by endoplasmic reticulum stress.It preliminarily revealed that the protective mechanism of Neuritin for the apoptosis induced by endoplasmic reticulum stress in the early brain injury,which provides a new theoretical basis for the treatment of SAH in the future.Methods:In this study we used ninety-six of healthy adult male SD rats which weight250~350g.According to the random principle,the experimental rats were divided into four groups:Control group,Sham group,SAH group,SAH+Neuritin.The rat SAH model was induced by injection of nonheparinized arterial blood into the prechiasmatic cistern.HE staining was used to identify the model successfully made.The rats were graded by the Garcia Neurobehavioral Ncore.The protein expression of GRP78,CHOP,Caspase-12,ASK1 and MMP9 were detected by Western Blot.The changes of blood-brain barrier were evaluated by using Evans blue staining method.The changes of cerebral edema in rats were evaluated by means of dry and wet weight of brain.Apoptosis of neurons in rat cortex was detected by TUNEL technique.Results:(1)Neurobehavioral scores were observed after SAH,whose value is significantly rose.The number of TUNEL cell was decreased noticeably.The expressions of GRP78?CHOP?Caspase-12 ?ASK1 were significantly increased.Olso EBI may cause the damage of blood brain barrier(BBB)and obviously increased brain water content(2)After the intervention of Neuritin,the neurobehavioral score increased,and the number of apoptosis decreased significantly.The expression of GRP78,CHOP,caspase-12 and ASK1 decreased.The outcome of Cerebral edema and blood-brain barrier permeability improved significantly.Conclusion:(1)The cell apoptosis mediated by endoplasmic reticulum stress was involved in the process of early brain injury.(2)Neuritin protects the early brain injury after subarachnoid hemorrhage,which may inhibit cell apoptosis and improve the permeability of blood-brain barr.
