Objective To evaluate the effect of different loading doses of dexmedetomidine on bispectral index?BIS?of propofol given by target-controlled infusion..Methods Forty ASA physical status?or?patients,aged 2060yr,weighing4480 kg,,scheduled for elective laparoscopic operation,were equally and randomly divided into3groups using a random number table:control group?C group?,dexmedetomidine group??group D1?and dexmedetomidine group??group D2?.Patients in the group D1,group D2 and group C were respectively administered with dixmedetomidine 0.3ug/kg,0.6ug/kg,or sodium chloride injection by micro-pumps in 10 minutes before anesthesia induction.All patients received target-controled infusion of propofol.The initial target concentration were 0.5?0.3?0.2?g/ml The plasma target concentration of propofol were adjusted according to the improved OAA/S score.The BIS and the concentration of propofol effect chamber were recorded before induction of the medical?T0?,and after the dexmedetomidine or sodium chloride infusion?T1?,and when OAA/S score were stable at4,3,2 and 1.We also recorded the number of intraoperative adverse events.All the data were used SPSS 19.0 statistical software.Result Age,body mass index have no significant difference among three group?P>0.05?Before taking the medication,the OAA/S of all groups were 5 and the BIS were more than 94.After dexmedetomidine loading dose were given,There were two patients'OAA/S were dropped to 4 and BIS felled to the range of 84 to 77 in group D1;and there were five patients in group D2.When the OAA/S was 5,4,3,2 and 1,the BIS of group D2 were lower than group C significantly?P<0.05?.When the OAA/S was 4,3and 2,the BIS of group D2 were also lower than group C.and the BIS of group D1 were lower than group D2.When the OAA/S was 4,3 and 2,the concentration of propofol effect chamber of groupD1 and group D2 were lower than group C,and the group D2were lower than group D2.Conclusion Compared with using propofol alone,Dexmedetomidine can reduce the BIS when OAA/S score were at same time.As the dose increased,the BIS value reduce more significant.Dexmedetomidine can reduce the dosage of propofol when combined with propofol and enhance the sedative effect of propofol.
