The Association Study Of Galanin Gene Polymorphism And Obsessive-Compulsive Disorder In The Chinese Han Population

Objective:To investigate the possible relationship between the SNPs of Galanin gene and Obsessive-compulsive disorder in Chinese Han population.Methods:All the elbows of the subjects who are in accordance with the study are venous blood(adding ethylenediamine tetra acetic acid anticoagulant),6m L per person,and the treated blood samples are stored in the refrigerator at low temperature(temperature-20?),and then DNA is extracted.Finally,the extracted DNA products are stored at low temperature of the refrigerator(-20?).We assessed and analysised the genotype and allele frequencies of the rs694066 locus in the galanin receptor gene of 400Obsessive-compulsive patients complying with the Diagnostic and Statistical Manual of Mental Disorders(DSM-5)and we have 459 healthy contols.The experimental samples that need to be tested are evenly spread on the MassARRAY SpectroCHIP chip,and then carefully tested in the MassARRAY Analyzer Compact mass spectrometer,and the results are analyzed with TYPER to obtain the required data.Then,determinening what is the relationship between the rs694066 and Obsessive-compulsive disorder.Sort out all the data and analyze and integrate the data with SPSS17.0 software.Using t test to analyze the sample data of the measurement.Chi square test is used to count the sample data that needed to be counted(the test standard is alpha=0.05).SHEsis software is used to analyze the data of genotypes and alleles.All the samples are stratified according to the age of onset,different symptom types and gender differences to further explore whether the distribution of allele frequency and rs694066 locus in each group is related to Obsessive-compulsive disorder.Results:The SNP detection rate of galanin rs694066 gene is 99.79%,with two alleles of A and G,three genotypes of AA,AG and GG.The genotype distribution of rs694066 in the obsessive-compulsive disorder group is in good agreement with the expected value(c~2=0.189,DF=1,P=0.664).In healthy controls,the observed values and expectations are well matched(c~2=4.668,DF=1,P=0.307).This indicates that the distribution of the two genotypes is consistent with the Hardy Weinberg equilibrium law,and there is no deviation in the selection of the samples.There is no significant difference in the distribution of galloctin gene rs694066 alleles between the obsessive-compulsive disorder group and the healthy control group(c~2=0.159,DF=1,P=0.690).There is no significant difference in genotype frequencies between the rs694066 locus of the obsessive-compulsive disorder group and the healthy control group(c~2=1.430,DF=2,P=0.489),too.There is no significant difference in the distribution of alleles and genotype frequencies between different gender and age groups(P>0.05).Conclusion:There is no correlation between SNPs rs894066 and Obsessive-compulsive disorder in Chinese Han population,which is not marked in Obsessive-compulsive disorder probably.
