BACKGROUND: the origin of progenitor cells and formation of bone,muscle,blood,lymph and other connective tissue mesoderm mesenchymal said(interstitial card 's cells of Cajal,ICC)and consisted of spindle without differentiation or more potential or energy or epithelioid cells of gastrointestinal mesenchymal tumors called gastric stromal tumor,about 60-70% of the gastrointestinal stromal tumor is gastric stromal tumor,accounting for all the gastric cancer in 2%,it has the tendency to a fixed direction to the differentiation and malignant development,its expected treatment effect mainly depends on the early discovery and timely treatment actively.At present,gastric stromal tumor preoperative diagnosis is still mainly rely on endoscopic ultrasound and CT examination,inappropriate preoperative biopsy may cause tumor rupture and hemorrhage,exacerbate the risk of tumor spread outward,so do not recommend routine preoperative biopsy or biopsy,the final diagnosis depends on pathological examination after operation,and with immunohistochemical staining,some diagnosis of difficult cases,there is no need to detect the mutation of c-kit or PDGFRA gene.In the previous study,researchers in this group have used gene chip technology to screen the major differentially expressed genes in gastric stromal tumors.According to the difference of gene screening standard Q-value Fold less than or equal to 5% and Change is greater than2 or less than 0.5,6 pairs of samples selected differentially expressed genes with a total of 3293,which belongs to the 2588 up-regulated genes,plays the role of tumor suppressor 705.Objective: to verify the expression of ETV1,DPP10,DKK4 and CXCL14 in gastric myometrial stromal tumors and adjacent normal tissues,and analyze their relationship with clinicopathological parameters of gastric muscularis propria.Methods: 12 cases of gastric muscularis propria experimental method for the selection of the Yantai Yuhuangding Hospital from July 2016 to July 2017 1 years application of endoscopic treatment of stromal tumors and peritumoral tissues using gene chip technique to screen out the main differences in the expression of genes,and the selection of ETV1,DKK4 two gene by reverse transcription and QPCR Technology: the prevalent methods of the existing real time quantitative PCR analysis of experimental data,divided into two types,one is the absolute quantification,the other is a relative quantitative.Absolute quantification,using standard curve to calculate the initial copy number of template;the relative quantitative method,is through the comparison of two kinds of samples,one is the treated samples,the other is without sample target transcripts processing sample or reference,the expression difference between the two.The definition of baseline is: in the PCR amplification reaction,in the first few cycles,the fluorescence signal has little change and is close to a straight line.Such a straight line is called the baseline.The definition of fluorescence threshold is based on the curve of fluorescence amplification,a value set artificially,which can be set at any position in the stage of fluorescence signal index amplification.But it is usually within 3 to 15 cycles in the setting of the usual fluorescence threshold.CT value: the number of cycles experienced in each reaction tube when the signal of the fluorescence reaches a set threshold.The CT value of each template has a linear relationship with the logarithm ofthe number of copies of the initial copy of the template.The more initial copies,the smaller the CT value,and vice versa.The standard curve can be drawn from the standard number of the initial copy number which has been known.The meaning of the ordinate is CT value.The meaning of the abscissa represents the logarithm of the initial copy number.Therefore,to calculate the initial copy number of the sample on the standard curve,only the CT value of the unknown sample is needed.2-Delta CT method is a simple method of real-time quantitative PCR experiments in the analysis of gene expression in many ways relative changes,which is a simple method for relative quantification,verify.Results:in 12 cases of gastric myoma,ETV1 and DKK4 were specifically expressed in the tumor tissue but were not expressed in the peritumoral tissue.Conclusion: ETV1 and DKK4 are specifically expressed in the pathogenesis of gastric stromal tumors.It is hopeful that future gastric stromal tumors can be detected by gene detection and provide new targets for targeted therapy of gastric stromal tumors.
