| Objective The C5L2 gene is located on the long arm of chromosome 19 and consists of337 amino acids.The mutation at 698C>T site causes the amino acid encoded by the 233 codon change from valine to leucine,and the mutation at 901G>T site caused the arginine encoded by the 300 th codon to be replaced by glutamine.C5L2 is a G protein-coupled receptor and is a functional receptor for acylating proteins.Acylation-stimul-ating protein can bind to C5L2 and become true functional receptors.Asp-C5L2 mediates glucose transport and regulates lipid metabolism and storage.In addition,C5L2 was also thought to be a dumb receptor for C5 a.In the advanced stage of atherosclerosis,high expression of C5L2 is directly related to high levels of pro-inflammatory cytokines.The aim of this study was to investigate the polymorphisms of SNPs C698 T and G901 A in the C5L2 gene and their association with large-artery atherosclerosis stroke.Methods In this study,whole blood was collected for DNA extraction and PCR-gene fragment polymorphism was used to genotype 254 patients with large atherosclerotic cerebral infarction and 228 controls for basal C698 T and G901 A gene loci.First,the DNA sample was extracted from the whole blood sample of the study subject,then the PCR amplification technology was used to achieve the in vitro amplification of the target DNA,and the appropriate DNA restriction endonuclease was selected according to the base sequence of the target gene,and the different genotypes were cut by enzyme digestion.The target gene was cut into different fragments of different lengths.Finally,the number and size of the digested fragments were observed by agarose gel electrophoresis to determine the genotype.Results(1)For the C698 T locus,the frequency of CT genotyping was 40(15.75%)in the case group and 16(7.02%)in the control group.The difference was significant(OR=2.477,95% CI=1.346~4.558,P=0.003);(2)For the G901 A locus,the AG genotype carrying frequency was 14(5.51%)in the case group and 19(8.33%)in the control group.Chi-square test showed no significant difference(OR=0.642,95% CI=0.314-1.311,P=0.221);(3)Using non-conditional logistic regression analysis to adjust body mass index,waist-to-hip ratio,and low-density lipoprotein After the confounding factors such as history of hypertension,history of diabetes,history of smoking,and other confounding factors,differences in CT genotype frequencies between the two groups were significant(OR=2.520,95%CI=1.076-5.317,P=0.033);(4)The interaction analysis showed that there was no interaction between C698 T site and smoking and hypertension(95% CI of relative excess risk ratio include 0).Conclusions(1)C5L2 gene C698 T locus is associated with the large-artery atherosclerosis stroke and is a risk factor for large-artery atherosclerosis stroke;(2)G901A locus and large-artery atherosclerosis stroke unrelated;(3)There is no interaction between C698 T locus and smoking and hypertension. |
