With the increase in human eating habits(excessive energy intake)and lifestyle(more static life),the incidence of obesity is increasing year by year,especially adolescent obesity has become a serious public health problem.Obesity can lead to diabetes,cardiovascular disease and high blood pressure and other chronic diseases,to the community to bring great pressure and heavy burden.The results of clinical studies show that obesity is a complex disease,not a single factor,and the study of gene and environment interaction is conducive to understanding the causes of obesity,which is conducive to avoid risks and reduce the incidence of obesity.Objective:Based on the survey of 672 physical indicators of 7 ~ 18 years old students in Qufu City,gene screening,questionnaire survey of diet and living habits,the polymorphism of MC4 R gene and the effect of daily diet and exercise habit on obese subjects were investigated.To provide a theoretical basis for the prevention of obesity.Methods:Based on 672 7 ~ 18 years old students in Qufu City,the subjects were divided into control group(441 persons)and obese group(231 persons)according to the classification criteria of overweight and obesity BMI of Chinese schoolchildren and adolescents.The body height,body weight,waist circumference,hip circumference,waist-to-hip ratio,body mass index(BMI)were calculated for the two groups.The time,type,type and frequency of drink drink,time,mode,intensity and frequency of the students were counted by questionnaires.The genotipe of MC4 R was assessed by both polymerase chain reaction(PCR)and single-strand conformation polymorphism(SSCP).The polymerase chain reaction products were sequenced directly to identify MC4 R gene sequencing.The effects of genotype and lifestyle on obesity were performed after obtaining informed consent.Results:1.There was no significant difference in sex ratio and mean age between the control group and the obese group(p(29)0.05).The height,weight,waist circumference,waist-to-hip ratio and BMI were significantly different(p(27)0.05).2.Of the 672 children enrolled in the study,425 children were wild-homozygous(GG),accounting for 63.2% of the total number of included studies.The genotype of 213 children was mutated heterozygous(GA),accounting for the total number of studies31.7%,and the genotype of 34 children was mutated homozygous(AA),accounting for5.01% of the total number of included studies.3.There was no significant difference between GG genotype and GA / AA genotype in control group and obese group(p(29)0.05).4.There was no significant difference between the control group and the obese subjects in the consumption of western fast food,fat and sweat,carbonated beverage and physical exercise and static life(P> 0.05).5.MC4 R rs12970134 gene GA / AA mutant and static life in the occurrence of childhood obesity(p = 0.023,OR = 1.68,95% CI 1.12 ~ 3.42).Conclusions:Static life may increase the risk of obesity by affecting the polymorphism of the MC4 R gene.It is important to reduce the risk of obesity by changing the lifestyle lifestyle for obese children.The future also need a larger number of samples to verify the interaction between genes and illusions on the effects of obesity in children,to provide theoretical support for the prevention of obesity.
