Abstract/Summary:
Background: Non-alcoholic fatty liver disease(NAFLD)is characterized by the accumulation of triglycerides and free cholesterol in the liver,which is the main manifestation of the metabolic syndrome in liver.The prevalence of NAFLD in general population is 20%-30%,and the prevalence could research 70%-90% in the obese or diabetic patients.Simple steatosis as the early stage of NAFLD,is characterized by the fatty degeneration or lipid deposition in liver cells.Simple steatosis could progress to the non-alcoholic steatohepatitis,fibrosis,and cirrhosis,even the hepatocellular carcinoma(HCC).The pathogenesis of NAFLD remain unclear,abundant reports shown that NAFLD may be associated with some factors of genetic,environmental,metabolic or some that are uncleared,among these factors,genetic factors are very important.LIPA gene encodes the lysosomal acid lipase(LAL),which could protect liver from the fat accumulation by hydrolysate cholesterol and triglyceride to free cholesterol,glycerin,and free fatty acids in lysosomes.Previous studies shown that single nucleotide polymorphisms of LIPA could result in the disorder of cholesterol metabolism.Objective: The study was conducted to investigate the effect of LIPA rs 1051338 、rs1412444 polymorphisms on the levels of serum lipids,and the association with the risk of NAFLD.Methods: A case-control study was conducted based on the Han population in Qingdao district.From January 1 to December 31 in 2017,400 subjects were selected from the Qingdao Municipal Hospital,which containing 200 ultrasonic diagnosed NAFLD patients(120 males,80 females)and 200 health controls(110 males,90 females).The blood samples of all the subjects were collected,the biochemical analyses(including ALT,AST,GGT,ALP,FPG,TG,TC,LDL,HDL,and TIBL)and the SNPs analyses of LIPA were conducted.The data analyses were conducted by the SPSS 24 software.Hardy-Weinberg equilibrium(H-W)method was used to analysis the represent of the genotype of the case group and the control group.Pearson chi-square and t-test were used for counting data and measurement data respectively.Odds ratio and 95% CI were calculated by logistic regression analysis.Results: No significant difference was observed of sex and age in the case group and health control(P > 0.05),there was a significant difference in BMI,SBP and DBP between the case group and health control(P < 0.01).The levels of serum ALT,AST,GGT,ALP,FPG,TG,TC,LDL and TIBL in the NAFLD group were all higher than those in control group(P < 0.05),and the levels of serum HDL were lower than control group(P < 0.05).There were no significant difference of the genotypes and alleles of LIPA rs1051338 and rs 1412444 in the NAFLD group and health control(P > 0.05).The serum TG、TC、LDL levels in rs 1051338 TT genotypes carriers were significant lower than GG/GT genotypes carriers in all series(P < 0.001,P < 0.001,P = 0.01).Conclusion: There were no significant association of the LIPA rs 1051338 、1412444polymorphisms with the risk of NAFLD in the Chinese Han population.The serum TG、TC、LDL levels in rs 1051338 TT genotypes carriers were significant lower than GG/GT genotypes carriers in all series.
