Objective :With the prevalence of type 2 diabetes mellitus(T2DM)and the aggravation of aging society,the number of cognitive impairment induced by it will increase.Therefore,some scholars have suggested that severe cognitive impairment may be a new long-term complication of T2 DM.Alzheimer’s disease(AD)is a common cognitive disorder.In view of the lack of effective treatment for AD,it is urgent to detect T2 DM patients with dementia at an early stage.So,we hope to find a reference for the diagnosis of AD.To seek whether there has common pathogenesis between AD and T2 DM.Try to provide a new breakthrough point for the study of comorbidity between AD and T2 DM.Methods :The blood specimen obtained from 20 healthy individuals and 20 elderly patients with T2 DM and 22 patients with AD and 20 patients with diabetic nephropathy(DN)and 20 people who had not only T2 DM but also AD.In this experiment,subjects selected from Qingdao municipal hospital from November 1,2016 to October 31,2017.ELISA method was used to detect the concentration of osteopontin(OPN),in peripheral venous blood samples of subjects.The concentration of ceruloplasmin(Cp)and beta-2-microglobulin(β2-MG)were measured by radioimmunoassay.Data were analyzed with SPSS 24.0 and draw the receiver operating characteristic curve(ROC curve).Results :1.There was no significant difference between the five groups of Cp(P=0.346).2.The content of β2-MG are different in the five groups(P=0.000).There were significantly higher in groups of AD,DN and T2DM+AD than those control group(P<0.005).Significant difference were found in groups of AD and T2DM+AD than T2 DM group(P<0.005).3.The level of OPN are different in the five groups(P=0.000).Significantdifference were found in groups of AD and T2DM+AD than T2 DM group(P<0.005).There were significantly higher in groups of AD and T2DM+AD than T2 DM group(P<0.005).4.The validity of OPN screening for AD is better.The receiver operating characteristic curve(ROC curve)was used to detect the reliability.The reliability of beta 2-MG for screening AD is high(the area under the curve is 0.719),and the OPN is higher(the area under the curve is 0.927).Conclusions:1.The generation of β2-MG and OPN in patients with AD were increased.And it may provide reference for the diagnosis of AD.2.The risk of AD increases when T2 DM patients with renal complications.Assessment of renal lesions can be used to screen for neurodegenerative diseases. |
