Objective To investigate the effects of fucoidan on blood lipid regulation and antioxidation of mice with apolipoprotein gene deficiency.Methods The healthy male Kunming mice 10 cases and absence of ApoE gene mice 30 cases were chosen and randomly divided into 4 groups: blank control group,Fucoidan treatment group,AS group,the negative control group.The effects of Fucoidan on lipid metabolism and AS plaques in model animals were observed by using ELASA kit to detect blood biochemical indexes in the peripheral blood of the AS model.Using HE staining,oil red O staining,polymerase chain reaction and immunohistochemistry methods to detect the expression of vascular endothelial growth factor(VEGF)and fibroblast growth factor-2(FGF-2)in oxidative stress signal pathway.Results The blank control group,Fucoidan treatment group,AS model group,the negative control group 4 group animal blood biochemical indexes in peripheral blood,in the comparison of Fucoidan treatment group compared with the AS model control group significantly decreased the total cholesterol,triglyceride and other indexes,statistically significant(P<0.05);low density lipoprotein cholesterol levels were significantly elevated,statistically significant difference(P<0.05).HE staining and oil red O staining showed that the size of atherosclerotic plaques in the Fucoidan treatment group was significantly smaller than that of the AS model group,and the difference was statistically significant(P<0.05).Immunohistochemistry showed that the expression level of VEGF in oxidative stress signaling pathway in Fucoidan treatment group was significantly higher than that in AS model group,and the difference was statistically significant compared with AS model group(P<0.05).The results of RT-PCR and western blot showed that the expression of VEGF and FGF-2 protein in the as model group wassignificantly higher than that in the blank control group(P<0.05).The expression of VEGF and FGF-2 protein in aorta of fucoid an treatment group was significantly different from that of the control group(P>0.05).Conclusion Fucoidan in brown fat can be significantly lower total cholesterol and triglyceride levels in the peripheral blood,improve the level of HDL cholesterol,reduce the formation of atherosclerotic plaque,enhanced oxidative stress signaling pathways and fibroblast growth factor-2 in the related factor expression,for prevention and treatment of atherosclerosis plays a positive role.
